摘要
为了提高角鲨烯的稳定性,以明胶和阿拉伯胶为壁材、角鲨烯为芯材、转谷氨酰胺酶为固化剂,采用复合凝聚法制备角鲨烯微胶囊。研究了壁材组成、芯壁比、壁材浓度、p H值、搅拌速度、乳化速度等单因素对微胶囊的包埋率、载药量以及粒径大小的影响,并通过正交试验优化了制备工艺。结果表明,角鲨烯微胶囊的最佳制备工艺:明胶:阿拉伯胶(w:w)为1.00:1.00,芯壁比为1:1,壁材浓度为2%,p H为3.6,搅拌速度为500 r/min,乳化速度10 000 r/min。该条件下,角鲨烯微胶囊的包埋率为(68.2±1.5)%,载药量为(40.8±1.2)%,粒径为(98.5±2.5)μm。
In order to improve the stability of squalene,microcapsules of squalene were prepared by complex coacervation using gelatin/arabic gum as the capsule wall,he squalene as the core material and transglutaminase as the curing agent.The effects of single factors such as wall material composition,core/wall ratio,wall material concentration,pH value,stirring speed and emulsifying speed,etc.on the encapsulation efficiency,drug-loading rate and particle size of microcapsules were investigated and the preparation process was optimized by orthogonal experiment.The results show that the optimal preparation process of squalene microcapsules was as follows:1.00:1.00 gelatin/arabic gum ratio,1:1 core/wall ratio,2%wall material concentration,3.6 pH value,500 rpm stirring speed and 10 000 rmp emulsifying speed.Under this condition,the encapsulation efficiency,drug-loading rate and particle size of microcapsules were(68.2±1.5)%,(40.8±1.2)%and(98.5±2.5)μm,respectively.
