摘要
目的探讨以大黄素为声敏剂介导声动力疗法对小鼠动脉粥样硬化治疗作用。方法高脂饮食诱导Apo E^(-/-)小鼠发生动脉粥样硬化。将50只Apo E^(-/-)小鼠分为不做任何处理的模型对照组、以5-氨基酮戊酸介导的阳性对照组、大黄素高、中、低剂量介导SDT组,每组10只。除模型组外,其余各组自高脂4周起,分别进行相应的声动力治疗,隔天1次,于12周处死小鼠。取心脏,制备切片,通过油红染色检测斑块大小。取主动脉,进行荧光定量PCR检测基质金属蛋白酶TMP-2、TMP-9和TIMP-1,分析对斑块稳定性的影响。同样检测细胞因子TNF-α、IFN-γ、IL-10和TGF-β,分析对炎症的影响。结果大黄素介导声动力疗法能减轻动脉粥样硬化斑块大小;显著增加TIMP-1而降低TMP-2和TMP-9在主动脉的表达。显著降低促炎性细胞因子TNF-α和IFN-γ而增加抗炎性细胞因子IL-10和TGF-β在主动脉局部的表达。结论大黄素介导声动力疗法可能通过降低炎症反应发挥抗动脉粥样硬化作用,该方法对于动脉粥样硬化的治疗具有积极意义。
Objective To investigate the therapeutic effect of emodin as a sonodynamic therapy on atherosclerosis in mice.Methods High fat diet induced atherosclerosis in ApoE-/-mice.50 ApoE-/-mice were divided into a model control group without any treatment,a 5-positive control group mediated by aminolevulinic acid,a large emodin,a medium and low dose SDT group,10 rats in each group.In addition to the model group,other groups were given fat 4 weeks,respectively,sonodynamic treatment,the next day 1,mice were sacrificed at 12 weeks.The heart was taken,the slices were prepared,and the size of the plaque was detected by oil and red staining.The aorta was taken and the matrix metalloproteinases TMP-2,TMP-9 and TIMP-1 were detected by fluorescence quantitative PCR,and the effect of the analysis on the stability of plaque was analyzed.The effects of cytokine TNF-α,IFN-gamma,IL-10 and TGF-βon the inflammation were also detected.Results Rhubarb mediated photodynamic therapy could reduce the size of atherosclerotic plaque,significantly increase TIMP-1 and reduce the expression of TMP-2 and TMP-9 in the aorta.The expression of anti-inflammatory cytokine TNF-αand IFN-gamma was significantly increased and the expression of anti-inflammatory cytokine IL-10 and TGF-βin the aorta was increased.Conclusion Emodin-mediated sonotherapy may play an anti-atherosclerotic role by reducing inflammatory response,which has a positive significance for the treatment of atherosclerosis.
