摘要
目的探讨脊髓CXCL13在瑞芬太尼诱发痛觉过敏中的机制。方法成年健康SD大鼠(SPF级)20只,随机分为4组:切口痛组(C组),瑞芬太尼组(R组),NC-si RNA慢病毒组(NC组)和CXCL13-si RNA慢病毒组(CX组)。大鼠行足跖底切口术后,I组:尾静脉泵注生理盐水10μg/(kg·min),持续1 h;R组:尾静脉泵注盐酸瑞芬太尼注射液10μg/(kg·min),持续1 h;NC组鞘内注射NC-si RNA慢病毒10μL(108TU/m L)半小时后,尾静脉泵注瑞芬太尼10μg/(kg·min),持续1 h;CX组:鞘内注射CXCL13-si RNA慢病毒10μL(108TU/m L)30 min后,尾静脉泵注瑞芬太尼10μg/(kg·min),持续1 h。采用von Frey法检测CXCL13敲除对瑞芬太尼诱发痛觉过敏的影响,采用免疫荧光双标法,Westernblotting,RT-PCR法观察瑞芬太尼诱发痛觉过敏进程中CXCL13对小胶质细胞的激活情况。结果与I组比较,R组、NC组术后6 h机械痛阈值随时间增加显著降低;而CX组较R组、NC组术后6 h机械痛阈值显著升高;R组、NC组较I组、CX组CXCL13表达增高,且与小胶质细胞的激活正相关。结论瑞芬太尼诱发痛觉过敏的机制可能与CXCL13激活小胶质细胞有关。
Objective To investigate the role of CXCL13 activation in the spinal cord in remifentanil?induced hyperalgesia.Methods Twenty rats were randomly divided into four groups:the incision pain after operation group(group I),the remifentanil group(group R),the NC?siRNA lentivirus group(group NC)and the CXCL13?siRNA lentivirus group(group NC).After the plantar incision surgery was performed in rats,saline was given by the continuous infusion in the caudal vein at the rate of 10μg/(kg·min)for 1 hour in group I.Remifent?anil was given by continuous infusion in the caudal vein at the rate of 10μg/(kg·min)for 1 hour in group R.After NC?siRNA lentivirus was given intrathecally for half an hour,remifentanil was given by continuous infusion in the caudal vein at the rate of 10 ug/kg.min for 1 hour in group NC and in group CX.Von Frey test was selected to assess the effect of CXCL13 knockdown on remifentanil?induced hyperalgesia.To detect the activation of microglial cells,expression of CXCL13 was detected by immunofluorescence,RT?PCR and Western blotting assay,respec?tively.Results Compared with group I,MWT was significantly lowered after incision in group R and group NC,but MWT after incision in group CX was significantly higher than that in group R and group NC.Compared with group I and group CX,CXCL13 expression in group R and group NC was significantly increased,with the positive correlation with the activation of microglia cells.Conclusion The activation of microglia cells by CXCL13 may be involved in the mechanism of remifentanil?induced hyperalgesia.
