一种嗜蓝孢孔菌新种的分离鉴定与菌株初步筛选 被引量:5

摘要 [目的]在山西同朔地区沙棘木上采集到野生嗜蓝孢孔菌属一新菌(Fomitiporia yanbeiensis),并筛选出适宜不同碳源的母种菌株株系及适宜不同主料产区的原种菌株株系,为母种和原种的生产提供理论依据。[方法]通过形态学特征、分子系统发育、组织分离培养等研究方法对该菌株进行分离纯化鉴定,并以葡萄糖、玉米粉、可溶性淀粉为碳源培养母种菌株,以小麦、玉米、小米为主料培养原种菌株。[结果]Fomitiporia yanbeiensis在3种碳源的母种培养基中均能生长,以玉米粉为碳源时菌丝平均生长速度最快,为2.22mm·d-1,其次是葡萄糖,为2.12mm·d-1,第三是可溶性淀粉,为2.05mm·d-1。其中1号菌株在玉米粉培养基中菌丝生长速度最快(2.27mm·d-1)且菌丝长势最强(+++);在葡萄糖培养基中,5号菌丝生长速度最快(2.17mm·d-1)且菌丝长势最强(+++);而在可溶性淀粉培养基中,6号菌丝生长速度最快(2.12mm·d-1)且菌丝长势最强(+++)。在小麦、玉米和小米三种原种培养基中,接种后30d和45d时,满袋率及平均满袋率最高为玉米培养基,其次是小麦培养基,第三是小米培养基。在玉米培养基中,1号菌株的满袋率最高,而小麦和小米培养基中,6号的满袋率最高。[结论]Fomitiporia yanbeiensis可确定为嗜蓝孢孔菌属的一个新种。筛选出适宜玉米粉培养的母种菌株为1号,适宜可溶性淀粉培养的为6号菌株,适宜葡萄糖培养的为5号菌株。适宜玉米培养的原种菌株为1号,适宜小麦和小米培养的均为6号菌株。 [Objective]A new species of Fomitiporia namely Fomitiporia sp.nov.was collected from the wild sea buckthorn shrubs in Datong and Shuozhou,Shanxi province.The suitable culture media were determined for both mother strains and original strains.The study provided theoretical basis for the production of both mother and original strains.[Methods]The strain was isolated and identified based on morphological characteristics,molecular phylogeny,and tissue isolation and culture.The mother strains were cultivated using glucose,corn flour,and soluble starch as different carbon sources,while wheat,corn,and millet were used for the original strains.[Results]Fomitiporiayanbeiensis was able to grow in mother culture media with all three carbon sources,and cultivation on corn medium resulted in faster mycelial growth at 2.22 mm·d^-1 as compared to glucose and soluble starch at 2.12 mm·d^-1 and 2.05 mm·d^-1,respectively.The highest mycelial growth rate was observed in No.1 strain at 2.27 mm·d^-1 and the strongest mycelial growth vigor was shown in corn flour medium.In glucose medium,No.5 mycelia grew fast at 2.17 mm·d^-1 and mycelial growth vigor was the strongest.In soluble starch medium,No.6 mycelia grew rapidly at 2.12 mm·d^-1 and mycelial growth vigor was the strongest.With respect to fructification,cultures on three original media showed the production rate from high to low as the order of maize medium,wheat medium,and millet medium in 30 and 45 days after inoculation,while No.1 strain performed the best in maize medium and No.6 strain in wheat medium and millet medium.[Conclusion]The present study provided solid evidence to regard Fomitiporiayanbeiensisas a new species of Fomitiporia genus.Cultivated in the mother culture,No.1 strain performed best with corn flour medium,while No.5 with glucose medium and No.6 with soluble starch medium.In the original culture,No.1 grew better with maize medium,and No.6 with both wheat and millet medium.
