Simultaneous Determination of Scopolamine and Dexmedetomidine in Blood Sample Using Ultra-performance Liquid Chromatographymass Spectrometry

摘要 目的建立血液中东莨菪碱和右美托咪定的液相色谱-串联质谱检测方法。方法样品经固相萃取、反相液相色谱分离后进行质谱检测,根据保留时间及特征离子进行定性,东莨菪碱以子离子m/z 137.9、右美托咪定以子离子m/z 95.0进行定量。结果东莨菪碱和右美托咪定均在1~200ng/m L范围内线性关系良好,平均回收率大于80%,日内、日间测定结果的相对标准偏差均小于10.4%,该方法两种药物的最低检出限均低于0.1ng/m L。结论该方法简单、灵敏,适用于同时检测血液中东莨菪碱和右美托咪定,能够满足利用东莨菪碱和右美托咪定杀人、抢劫、强奸等案件的检验鉴定。 Objective A simple,specific and sensitive method is to develop and validate for simultaneous determination of scopolamine and dexmedetomidine in blood sample by ultra performance liquid chromatography-mass spectrometry(UPLCMS/MS).Methods The blood sample was extracted with Oasis MCX and separated through reversed phase chromatographic column.Scopolamine and dexmedetomidine were qualitatively analyzed by each retention time and two ion pairs of precursor/product observed in multiple reaction monitoring(MRM),Mass spectrometry scan at 303.8/137.9 and 201.15/95.0 were used for quantitative analysis of scopolamine and dexmedetomidine,respectively.Results The calibration curve was linear within the range of 1-200ng/mL for scopolamine and dexmedetomidine.The method recovery was more than 80%.The RSD of inter and intra-day were both less than 10.4%.The limit of quantitation was 0.1ng/mL.Conclusions The method is accurate,specific,sensitive,and suitable for the determination of criminal case of murder,robbery and rape.
