摘要
为了比较4个罗氏沼虾抗病选育群体(A、B、C、D)的抗病性能,对4个罗氏沼虾选育群体和1个非选育群体(E)进行了溶藻弧菌肌肉注射感染试验,运用高通量测序方法比较不同罗氏沼虾群体间感染组和对照组肌肉组织中弧菌的变化情况,根据感染成活率的差异性,分析其中3个罗氏沼虾群体(A、C、E)肠道组织和养殖水体的菌群多样性。结果表明:肌肉组织样品中微生物主要分为6个门,其中厚壁菌门占绝对优势,次优势门为变形菌门,软壁菌门和酸杆菌门的含量相对较低;各感染组芽孢杆菌属、假单胞菌属、嗜冷杆菌属的含量都高于对照组,而乳球菌属、乳酸杆菌属的含量明显低于对照组;肠道组织和养殖水体中微生物主要分为3个门,分别是变形菌门、拟杆菌门和厚壁菌门;基于不同罗氏沼虾群体肌肉组织中不同时间段内溶藻弧菌的含量,5个群体的抗弧菌感染能力分别为A>B>E>D>C,与感染成活率结果一致。综上,罗氏沼虾体内的菌群结构和养殖水环境密切相关,但相对独立;芽孢杆菌和类芽孢杆菌的含量差异可能是造成A群体和C群体感染成活率差异显著的原因。
To further understand the disease resistance of four anti-disease breeding populations(A,B,C and D)and to provide theoretical basis for further breeding,four anti-disease breeding populations of Macrobrachium rosenbergii and non-resistant breeding population(E)were injected with Vibrio alginolyticus,and the control group of each population was infected with normal saline.High-through sequencing technique was used to compare the variations between test group and control group.Bacterial diversities in intestinal tissue and aquaculture water of three populations(A,C and E)were analyzed according to the difference of the infection survival rate.The results revealed that microorganisms in muscle tissues were mainly divided into six phyla,of which the dominant and the subdominant phyla were Firmicute and Proteobacteria,respectively,and then the contents of Tenericumes and Acidobacteria were relatively low.The contents of Bacillus,Pseudomonas and Psychrobacter in each test group were higher than those in each control group,however,the contents of Lactococcus and Lactobacillus were obviously lower than those of the control group.Microorganisms in intestinal tissues and aquaculture waters were mainly divided into three phyla,including Proteobacteria,Bacteroidetes and Firmicutes.According to the residue of Vibrio alginolyticus,the disease resistances in all five populations were A>B>E>D>C,consistent with the result of the infection survival rate.There existed a close correlation between bacterial species composition in the body of Macrobrachium rosenbergii and aquacultural environment.Meanwhile,the bacterial species composition in the body was relatively independent.Bacillus and Paenibacillus might be the cause of significant difference in the survival rate of A and C populations.
