摘要
目的检测RANK、RANKL、OPG蛋白在含牙囊肿、角化囊性瘤、单囊型成釉细胞瘤囊壁上皮层的表达,并比较目标蛋白表达水平在三组疾病差异。方法采用免疫组化(SP)染色检测口腔黏膜、含牙囊肿、牙源性角化囊性瘤、单囊型成釉细胞瘤囊壁上皮层中RANK、RANKL及OPG的表达。结果三组疾病样本中RANKL表达强度高于黏膜(P <0.05)但疾病组之间不存在差异;OPG表达强度依次为:正常黏膜>角化囊性瘤>单囊型成釉细胞瘤;RANK表达强度在各实验组之间不存在差异(P> 0.05)。结论在实验样本上皮层中均能检测到OPG、RANK、RANKL的表达;含牙囊肿、角化囊性瘤、单囊型成釉细胞瘤的颌骨吸收过程与RANKL高表达和OPG低表达密切相关,三组疾病骨吸收的差异可能与OPG的表达水平相关。
Objective To explore the expression of RANK,RANKL and OPG in dentigerous cysts(DC),keratocystic odontogenic tumors(KCOT)and unicystic ameloblastomas(UA).To compare the differences of expression of these genes in those diseases.Methods The immunohistochemical expression(SP way)of these biomarkers were evaluated in lining epithelium of DC KCOT and UA,compared with oral mucosa.Results The intensity of RANKL expression in disease groups were higher than that in mucosa(P<0.05),but there was no statistically significance among disease groups(P>0.05).OPG expression intensity showed mucosa>KCOT>UA.However,the intensity of RANK cannot be considered difference between the test groups.Conclusion The positive expression of OPG,RANK and RANKL was detected in the epithelium of four experimental groups.Our findings indicate that overexpression of RANKL and lower-expression of OPG are closely related with jaw bone resorption in UA,KCOT and DC.Furthermore,bone resorption differences of those diseases are relevant with intensity of OPG.
