摘要
目的研究二苯乙烯苷对人结肠癌细胞增殖及凋亡的作用,并探讨潜在的分子机制。方法通过CCK8检测二苯乙烯苷对细胞增殖及顺铂化疗增敏的影响;流式细胞仪检测二苯乙烯苷对细胞凋亡的影响,Western blot检测二苯乙烯苷处理细胞后PI3K/AKT通路相关指标的变化。结果 CCK8结果显示,THSG可抑制HCT116和SW480细胞增殖,其对细胞的抑制呈时间和剂量依赖。THSG与顺铂联合运用可增加HCT116和SW480细胞对顺铂的敏感性。流式细胞仪检测结果显示,THSG作用于HCT116和SW480细胞后,细胞早期凋亡率呈剂量依赖递增。Western blot结果显示,THSG可上调PTEN的表达,下调P-AKT、P-IKKβ的磷酸化水平。结论 THSG可能通过抑制PI3K/AKT/NF-κB通路从而抑制HCT116和SW480细胞增殖并促进凋亡。
Objective To identify the effect of 2,3,5,4′-Tetrahydroxy stilbene-2-O-β-D-glucoside on proliferation and apoptosis of colon cancer cells,and expore the relevant the underlying molecular mechanisms.Methods The effect of THSG on proliferation and enhance the sensitivity to cisplation of HCT116 and SW480 was evaluated by CCK8 assay,and the apoptotic rate of HCT116 and SW480 cells was investigated by flow cytometry.The expression of PTEN,p-AKT、AKT were examined by western blot.Results THSG could significantly decrease cell proliferation in HCT116 and SW480 in a time and dose dependent way.THSG and cisplatin working together on HCT116 and SW480 cells obviously increase the sensibility to cisplatin.And THSG can increase the apoptosis of HCT116 and SW480 cells in a dose dependent way.Western blot shows THSG can enhance the expression of PTEN and reduce the phosphorylation-level of AKT.Conclusion THSG inhibit the proliferation of HCT116 and SW480 cells and promote apoptosis may through depressing the PI3K/AKT/NF-κB signaling pathway.
