摘要
研究野生党参芦头和参尾的高效液相色谱(high performance liquid chromatography,HPLC)特征图谱,并对其含有的多种活性成分的含量进行比较,为指导党参芦头和参尾的合理利用提供依据。采用HPLC法对党参炔苷、苍术內酯Ⅲ及5-羟甲基糠醛的含量进行同时测定,采用苯酚-硫酸法测定党参多糖的含量,采用指纹图谱相似度评价系统(2008版)软件对野生党参芦头和参尾特征图谱进行分析。结果表明,野生党参芦头和参尾HPLC特征图谱的相似度达0.90以上,二者中主要成分均是多糖,参尾中的多糖、苍术內酯Ⅲ及5-羟甲基糠醛的含量远高于芦头,党参炔苷在芦头和参尾中的含量却相差不大。党参芦头和参尾的主要化学特征相似,芦头中含有的党参炔苷及主成分多糖具有很好的药用价值,仍然可以加以开发利用。
The aim is to establish the high performance liquid chromatography(HPLC)specific chromatograms for content determination of multiple components having bioactivities in rhizomes and sterns of wild Radix Codonopsis,which provided basis for rational utilization of the rhizomes and sterns of wild Radix Codonopsis.The HPLC was adopted to simultaneously quantify lobetyolin,atractylenoideⅢand 5-hydroxymethyl furfural.The content of polysaccharides was determined by phenol-sulfuric acid method.The HPLC specific chromatograms were analyzed by similarity evaluation system for chromatographic fingerprints of traditional Chinese medicine(TCM)(version 2008).The study showed the similarity in HPLC specific chromatograms between rhizomes and sterns of wild Radix Codonopsis was more than 0.90,and the main ingredient was polysaccharides,the contents of polysaccharides,atractylenoideⅢand 5-hydroxymethyl furfural were more abundant in sterns than in rhizomes,but the contents of lobetyolin were almost the same in rhizomes and sterns.The main chemical feature was similar in rhizomes and sterns of Radix Codonopsis,both the lobetyolin and the principal polysaccharides had good medicinal values.In the future,rhizomes of Radix Codonopsis could be exploited as a high-quality resource.
