摘要
目的初步探讨左、右半结肠癌基因转录水平的分子差异,及其差异表达基因INHBA与结肠癌临床病理特征及预后的相关性。方法收集具有完整随访资料的结肠癌标本,分析左、右半结肠癌患者的生存情况,同时利用公共数据库中的结肠癌数据,筛选左、右半结肠癌组织的差异表达基因,并应用RT-PCR及免疫组化分析差异表达基因INHBA的表达水平与结肠癌临床病理特征及预后的相关性。结果结直肠癌公共数据库分析结果显示,左、右半结肠癌转录组水平差异表达基因在TGF-β、NF-κB等信号通路中显著富集,TGF-β通路中的相关差异表达基因INHBA在右半结肠癌组织中的mRNA及蛋白表达水平显著高于左半结肠癌(P <0. 05),并且INHBA高表达与右半结肠癌的脉管浸润、远处转移及不良预后呈正相关(P <0. 05)。结论 INHBA基因在右半结肠癌组织中的表达水平显著高于左半结肠癌,并且其表达与患者的恶性进展及不良预后密切相关,INHBA表达上调可能在右半结肠癌的发生和演进过程中发挥着重要作用。
To investigate the molecular differences in the gene transcription levels of left and right colon cancers,and the relationship between the differentially expressed gene INHBA and the clinicopathological parameters and prognosis of colorectal cancer.Methods Specimens of colorectal cancer patients with complete follow-up data were collected to analyze the survival status of patients with left or right colon cancer and colorectal cancer data in public databases were used to screen differentially expressed genes in left and right colon cancer tissues.Also,real-time quantitative PCR and immunohistochemistry were used to analyze the relationship between the expression level of differentially expressed gene INHBA and the clinicopathological parameters and prognosis of patients with colorectal cancer.Results Analysis of public databases for colon cancer revealed that TGF-βand NF-κB signaling pathways were significantly enriched in the right colon cancer,and the mRNA and protein expression levels of the related differentially expressed genes INHBA in the TGF-βpathway in the right colon cancer were significantly higher than those of left colon cancer(P<0.05).Also,the high expression of INHBA was positively correlated with vascular infiltration,nerve invasion,distant metastasis and poor prognosis(P<0.05).Conclusion The expression level of INHBA gene in right colon cancer tissue is significantly higher than that in left colon cancer,and its expression is closely related to malignant progression and poor prognosis of tumor patients,suggesting that INHBA gene may play an important role in the oncogenesis and progression of right colon cancer.
