摘要
目的探讨日本血吸虫虫卵可溶性抗原(Sj SEA)对三硝基苯磺酸(TNBS)诱导的小鼠结肠炎的保护作用。方法将Balb/c雌鼠分为三组:正常对照组、TNBS模型组、TNBS+Sj SEA干预组(TNBS灌肠后6 h、24 h经腹腔注射给予50μg Sj SEA)。测定小鼠肠疾病活动指数(DAI)、体质量变化、结肠长度、宏观炎症评分(MAO)、镜下组织学评分(MIO)和髓过氧化物酶活性(MPO)及HE染色。同时用流式细胞仪(FACS)检测各组脾中Th1、Th2、Treg的表达。结果与TNBS模型组相比,TNBS+Sj SEA干预组小鼠体质量、结肠长度均有所回升,DAI降低,结肠组织MAO、MIO、MPO均显著降低;HE染色结果显示,TNBS+Sj SEA干预组小鼠结肠炎症程度减弱;FACS检测结果显示:Sj SEA干预后的小鼠脾Th1(CD4+IFNγ+)降低,Th2(CD4+IL-4+)、Treg显著升高。结论 Sj SEA可有效减轻TNBS诱导的小鼠结肠炎症,此保护作用可能与平衡T淋巴细胞亚群Th1/Th2和Treg细胞免疫相关因子的表达、下调Th1、上调Th2、升高CD4+CD25+Foxp3+Treg细胞的表达比例有关。
Objective To investigate the protective effect of Schistosoma japonicum soluble egg antigens on TNBS-induced colitis.Methods Three groups of female Balb/c mice were used:mice in the control group were treated with PBS(control-PBS),and TNBS colitis mice were treated with PBS(TNBS-PBS)and S.japonicum soluble egg antigens(TNBS-SjSEA)separately.Mice were treated with 50μg S.japonicum soluble egg antigens through intraperitoneal injection at 6 h and 24 h after TNBS-colitis induction.The pathological and inflammatory parameters were estimated by measuring weight loss,disease activity index(DAI),colon length,myeloperoxidase(MPO)activity,macroscopic score(MAO),microscopic score(MIO),and Histological examination.Th1,Th2 cells and Tregs were measured in spleens by flow cytometry so as to determine the S.japonicum soluble egg antigens induced immune response.Results The pathology and inflammatory parameters and expression of IFN-γin the spleens were significantly reduced after treatment with S.japonicum soluble egg antigens in colitis mice,whereas the production of IL-4 was relatively elevated and Tregs were lifted in the spleens.Conclusion S.japonicum soluble egg antigens may alleviate experimental colitis in mice via increasing Tregs and Th2 mediated response and,correspondingly,suppressing Th1 dominant response which is responsible for the pathogenesis of colitis.
