沉默FBI-1基因对乳腺癌MCF-7细胞周期和凋亡的影响 被引量:2

摘要 目的研究沉默FBI-1基因表达对乳腺癌MCF-7细胞周期和凋亡的影响,并探讨可能的作用机制。方法采用shRNA干扰技术沉默MCF-7细胞中FBI-1基因的表达,qRT-PCR及Western blot鉴定FBI-1基因的沉默效率。流式细胞术检测细胞周期和凋亡情况,qRT-PCR及Western blot检测p53和p21表达。结果经sh RNA干扰后FBI-1 mRNA和蛋白水平显著降低;FBI-1基因沉默后,G0/G1期细胞数明显增加,S期和G2/M期细胞数显著下降,细胞凋亡率显著增高,p53和p21 mRNA及蛋白表达明显上调。结论沉默FBI-1基因表达可诱导细胞G1/G2期阻滞和细胞凋亡,其机制可能通过上调p53和p21 mRNA及蛋白表达有关。 Objective To investigate the effects of FBI-1 silencing on cell cycle and apoptosis of breast cancer MCF-7 cells and to explore the possible mechanism.Methods RNA interference method was used to silence FBI-1 expression in MCF-7 cells.The silencing efficiency of FBI-1 gene was identifed by qRT-qPCR and Western blot assay,respectively.The changes of cell cycle distribution and the apoptosis rate were detected by flow cytometry.p53 and p21 expressions were detected by qRT-qPCR and Western blot assay,respectively.Results FBI-1 expression was efficiently decreased by FBI-1 shRNA.The percentage at G0/G1 phase was increased,the percentages at S and G0/G1 phase were decreased,and the apoptosis rate was increased in MCF-7 cells with FBI-1 silencing.However,expressions of p53 and p21 were upregulated.Conclusion FBI-1 silencing arrests the cell cycle at G0/G1 phase and induces apoptosis of MCF-7 cells,and the possible mechanism may be associated with-the up-regulation of p53 and p21 expressions.
