摘要 目的:将贴壁培养型BHK21细胞驯化成适合口蹄疫疫苗生产的无血清全悬浮培养型细胞.方法:引进贴壁培养型BHK21细胞,通过改变培养液和降低血清的方法进行悬浮培养的驯化,对驯化得到的悬浮培养型细胞进行生物学检定.结果:引进的BHK21细胞低血清驯化培养22代可悬浮生长,用无血清培养基培养10代后形态趋于稳定生长变快.贴壁和悬浮细胞均有致瘤性,对口蹄疫病毒敏感,适应生物反应器高密度培养,且没有外源物污染.结论:驯化的BHK21细胞可用于口蹄疫疫苗的研究和生产.
