摘要
目的研究多种耐药蛋白在三阴性乳腺癌中表达的临床意义。方法采用免疫组化法检测76例三阴性乳腺癌患者谷胱甘肽S转移酶(GSTπ)、P-糖蛋白(Pgp)、肺耐药蛋白(LRP)和拓扑异构酶-Ⅱ(TOPOⅡ)的表达,分析它们与患者的临床病理特征的相关性。结果三阴性乳腺癌患者中GSTπ、Pgp、LRP、TOPOⅡ的阳性表达率分别为63.16%、25.00%、34.21%、32.89%,GSTπ的高表达与脉管癌栓有相关性,而与患者年龄、肿瘤大小、组织学分级无相关性; TOPOⅡ的表达与患者的年龄有相关性,而与肿瘤大小、淋巴结转移、脉管癌栓及组织学分级无相关性; Pgp、LRP的表达与患者年龄、肿瘤大小、淋巴结转移、脉管癌栓和组织学分级均无相关性; GSTπ与LRP表达呈正相关(r=0.393,P=0.003),其他基因表达水平间无相关性。结论 GSTπ表达水平与三阴性乳腺癌临床病理特征如血道转移等相关,且GSTπ与LRP表达呈正相关,提示联合检测GSTπ和LRP在三阴性乳腺癌中的表达对判断肿瘤转移具有一定的参考价值。
Objective To study the clinical significance of expression of multidrug resistance protein in triple-negative breast cancer.Methods The expressions of glutathione S-transferase(GSTπ),P-glycoprotein(Pgp),lung resistant protein(LRP)and topoisomeraseⅡ(TOPOⅡ)in 76 patients with triple-negative breast cancer were detected by immunohistochemical method,and the correlation between them and clinicopathological features was analyzed.Results The positive expression rates of GSTπ,Pgp,LRP and TOPOⅡin patients with triple-negative breast cancer were 63.16%,25.00%,34.21%and 32.89%,respectively.The high expression of GSTπwas correlated with vascular tumor thrombus,but not with age,tumor size and histological grade.The expression of TOPOⅡwas correlated with age,but not with tumor size,lymph node metastasis,vascular tumor thrombus and histological grade.The expressions of Pgp and LRP were not correlated with age,tumor size,lymph node metastasis,vascular tumor thrombus and histological grade.There was positive correlation between the expressions of GSTπand LRP(r=0.393,P=0.003),but there was no correlation between other gene expression levels.Conclusion GSTπexpression level is associated with clinicopathological features of triple-negative breast cancer such as hematogenous metastasis,and GSTπand LRP expression are positively correlated.These results suggest that the combined detection of GSTπand LRP expressions in triple-negative breast cancer have certain reference value for judging tumor metastasis.
