摘要
目的:建立蒙药阿那日-4中鞣花酸和胡椒碱的含量测定方法。方法:采用Agilent XDB-C18(250mm×4. 6 mm,5μm)色谱柱,以甲醇-0. 1%磷酸水进行梯度洗脱,流速为1. 0 m L·min-1;柱温为30℃;检测波长为272 nm。结果:鞣花酸在0. 146~0. 730 mg·mL^(-1)范围内线性关系良好(r=0. 9996),胡椒碱在0. 090~0. 450 mg·mL^(-1)范围内线性关系良好(r=0. 9992);平均回收率分别为98. 95%(RSD为0. 82%,n=5)和96. 25%(RSD为1. 07%,n=5); 3批样品中鞣花酸和胡椒碱的平均含量分别为3. 26 mg. g-1和2. 18 mg. g-1。结论:建立的方法简便,准确,重复性好,可用于蒙药阿那日-4中鞣花酸和胡椒碱的含量测定。
Objective:To establish a method for the determination of ellagic acid and piperine in Mongolian medicine Anari-4.Methods:Chromatographic separation was carried out with Agilent XDB-C18(250mm×4.6mm,5μm)chromatographic column and gradient elution with methanol-0.1%phosphoric acid solution at a flow rate of 1.0 mL·min^-1.The column temperature was 30℃;the detection wavelength was 272 nm.Results:The linear range was 0.146-0.730 mg·mL^-1(r=0.9996)for ellagic acid,and 0.090-0.450mg·mL^-1(r=0.9992)for piperine.The average recoveries of these compounds were 98.95%(RSD=0.82%,n=5)and 96.25%(RSD=1.07%,n=5),respectively.The average contents of ellagic acid and piperine in three batches of commercially available samples were 3.26mg.g^-1 and 2.18mg.g^-1,respectively.Conclusion:The established HPLC method is accurate,convenient,reliable and suitable for the determination of the content of ellagic acid and piperine in Mongolian medicine Anari-4.
