摘要
目的探究复方丹参注射液对重症急性胰腺炎(SAP)肺损伤(ALI)大鼠肺组织的改善作用,并探究其可能的作用机制。方法将SD大鼠分为假手术组、模型组、复方丹参注射液(SM)低、中、高剂量组、地塞米松组,每组12只大鼠,HE染色观察胰腺、肺组织损伤情况;测量腹水量、肺组织干、湿质量;测定肺泡灌洗液(BAL)中中性粒细胞(PMN)数量以及动脉血中血气指标;蛋白免疫印迹(WB)、免疫组化法分析CYLD/NF-κB通路蛋白表达;酶联免疫吸附法检测BAL中炎性细胞因子水平。结果与假手术组比较,模型组大鼠胰腺、肺组织病理评分、腹水量、胰腺、肺组织W/D、PMN、PaCO2、p-p65、p-IκBα蛋白表达、TNF-α、IL-1β、IL-6、MDA升高,PaO2、OI、CYLD蛋白表达、SOD水平降低(P <0.05)。与模型组相比,SM组、地塞米松组胰腺、肺组织病理评分、腹水量、胰腺、肺组织W/D、PMN、PaCO2、p-p65、p-IκBα蛋白表达、TNF-α、IL-1β、IL-6、MDA均降低,PaO2、OI、CYLD蛋白表达、SOD水平均升高(P <0.05)。结论复方丹参注射液可缓解SAP-ALI大鼠炎症损伤,可能与激活肺组织CYLD、抑制/NF-κB信号通路激活有关。
Objective:To explore the improvement effect of Compound Danshen Injection on lung tissues of rats with severe acute pancreatitis(SAP)-acute lung injury(ALI),and to explore its possible mechanism.Methods:SD rats were divided into the sham operation group(sham),SAP-ALI group,low dose,middle dose and high dose Compound Danshen Injection(SM)groups,and Dexamethasone group(DXM),with12rats in each.The damages of pancreas and lung tissues was observed by HE staining;the amount of ascites,lung tissue dry and wet weights,the number of neutrophils(PMN)in the bronchoalveolar lavage fluid(BAL)and blood gas index in arterial blood were measured;Western blot(WB)and immunohistochemistry were used to analyze the protein expression of CYLD/NF-κB pathway,enzyme-linked immunosorbent assay to detect the levels of inflammatory cytokines in BAL.Results:Compared with the sham group,all indexes of SAP-ALI group were improved(P<0.05).Compared with the SAP-ALI group,the indexes of SM group and DXM group were improved(P<0.05).Conclusion:Compound Danshen Injection can alleviate inflammatory injury of SAP-ALI rats,which may be related to activation of CYLD in lung tissue and inhibition of activation of NF-κB signaling pathway.
