摘要
观察天麻钩藤饮对脑出血大鼠TLR4、NF-κB、IL-1β表达的影响,探讨其保护神经功能的可能机制。60只Wistar雄性大鼠随机分为假手术、模型组和天麻钩藤饮组,每组按造模后1、3、5、7d分为4个亚组,采用自体血注入法建立脑出血大鼠模型。天麻钩藤饮组予大鼠天麻钩藤饮(18g/kg·d)灌胃,模型组和假手术组予等量生理盐水(3mL/kg·d)灌胃,均1次/24h。分别于试验第1、3、5、7天进行神经功能缺损评定、测定脑组织水含量和血清TLR4、NF-κB、IL-1β表达的水平。结果表明,与假手术组相比,模型组各个时间节点神经功能缺损评分、脑组织含水量和血清TLR4、NF-κB、IL-1β表达水平明显升高(P<0.05);与模型组相比,天麻钩藤饮组各个时间节点经功能缺损评分、脑组织含水量和血清TLR4、NF-κB、IL-1β表达水平明显降低(P<0.05)。天麻钩藤饮保护脑出血大鼠的神经功能可能与其减轻脑水肿及抑制TLR4、NF-κB、IL-1β的表达有关。
To investigate the effects of Tianma Gouteng decoction on the expressions of serum TLR4,NF-κB,IL-1βin rats with intracerebral hemorrhage and explore its mechanism preliminary.60male Wistar rats were randomly divided into the sham operation control group,the model group and the Tianma Gouteng decoction group,and then further divided into four time-point subgroups(1,3,5,7days after modeling,n=5/subgroup).The ICH model was established by injection of the rat,s auto blood(50μl)into the putaman region.The rats in the Tianma Gouteng decoction group were given with Tianma Gouteng decoction by intragastric administration(18g/kg·d),and those in the sham operation control group and model group were given with physiological saline(3mL/kg·d).The contents of water of brain tissue,the neurological function score and serum TLR4,NF-κB,IL-1βat1,3,5,7d were tested.Compared with the sham operation control group,the neurological function score,the brain water content,the levels of serum TLR4,NF-κB,IL-1βof the rats in the model group were higher significantly(P<0.05);Compared with the model group,the neurological function score,the brain water content,the levels of serum TLR4,NF-κB,IL-1βof the rats in Tianma Gouteng decoction group were lower significantly(P<0.05).Tianma Gouteng decoction could reduce the levels of proinflammatory factors TLR4,NF-κB,IL-1βin the ICH rats with intracerebral hemorrhage,which may contribute to its effects in improving neurological function.
