丹参提取物对高血压性心脏病大鼠神经免疫的调节作用初探 被引量:8

摘要 目的:探讨丹参提取物对高血压性心脏病(HHD)大鼠神经免疫的调节作用。方法:构建HHD大鼠模型,随机分为A组、B组、C组和D组,每组10只,另取10只健康大鼠记为对照组,A组和对照组腹腔注射生理盐水5 ml,B组、C组和D组分别腹腔注射0. 75 ml/mg、1. 5 ml/mg、3. 0 ml/mg各5 ml丹参乙醇提取物溶液,注射持续4周。采用心脏超声诊断仪测定左心室收缩末期内径(LVESD)、舒张末期内径(LVEDD)、左心室射血分数(LVEF),采用尾动脉无创性血压测量收缩压(SBP)、舒张压(DBP);酶联免疫法测定外周血P物质(SP)、血管活性肠肽(VIP)、去甲肾上腺素(NE)、血管紧张素Ⅱ(AngⅡ)、白细胞介素10(IL-10)、肿瘤坏死因子α(TNF-α)水平,流式细胞术检测外周血自然杀伤性细胞(NK)、T淋巴细胞亚群(CD4+、CD8+)活性,计算CD4+/CD8+值。结果:五组所检测指标比较均有显著性差异(P<0. 05),A、B、C、D组LVEF、IL-10、SP、VIP、NK、CD4+/CD8+显著高于对照组(P<0. 05),LVEDD、LVESD、SBP、DBP、NE、AngⅡ、TNF-α显著低于对照组(P<0. 05);B、C、D组LVEF、IL-10、SP、VIP、NK、CD4+/CD8+显著高于A组(P<0. 05),LVEDD、LVESD、SBP、DBP、NE、AngⅡ、TNF-α显著低于A组,C、D组LVEF、IL-10、SP、VIP、NK、CD4+/CD8+显著高于B组,LVEDD、LVESD、SBP、DBP、NE、AngⅡ、TNF-α显著低于B组(P<0. 05);D组LVEF、IL-10、SP、VIP、NK、CD4+/CD8+显著高于C组(P<0. 05),LVEDD、LVESD、SBP、DBP、NE、AngⅡ显著低于C组(P<0. 05),C、D组TNF-α水平差异无统计学意义(P>0. 05)。结论:丹参提取物对HHD大鼠有改善心脏功能和降低血压作用,推测很可能通过调节神经免疫因子水平发挥作用。 Objective:To observe the effect of salvia miltiorrhiza extract on neuroimmunization of hypertensive heart disease rats.Methods:Construction of HHD rat model,and the HHD rats were randomly divided into the group A,group B,group C and group D,with10rats of each group,and another10healthy rats were taken as the control group.The group A and the control group were injected saline,and the group B,the group C and the group D were respectively injected0.75ml/mg,1.5ml/mg,3.0ml/mg of Salvia miltiorrhiza ethanol extract solution,all5ml of intraperitoneal,and the injection last for4weeks.The left ventricular end systolic diameter(LVESD),the end diastolic diameter(LVEDD)and the left ventricular ejection fraction(LVEF)were detected by cardiac ultrasonography.The systolic blood pressure(SBP)and the diastolic blood pressure(DBP)were detected by systolic blood pressure.The levels of the substance P(SP),the vasoactive intestinal peptide(VIP),the norepinephrine(NE),the angiotensinⅡ(AngⅡ),the interleukin10(IL-10)and the tumor necrosis factorα(TNF-α)of peripheral blood were detected by Enzyme-linked immunoassay.The activity of the natural killer cells(NK)and the T cell subset(CD4+,CD8+)of peripheral blood were tests by Flow cytometry,and the CD4+/CD8+values was calculated.Results:There were significant differences among the five groups(P<0.05),and the levels of LVEF,IL-10,SP,VIP,NK and CD4+/CD8+of the A,B,C and D groups were significantly higher than those of the control group(P<0.05),while the LVEDD,LVESD,SBP,DBP,NE,AngⅡand TNF-αwere significantly lower than those of the control group(P<0.05).The levels of LVEF,IL-10,SP,VIP,NK and CD4+/CD8+of the A,B,C and D groups were significantly higher than those of the A group(P<0.05),while the LVEDD,LVESD,SBP,DBP,NE,AngⅡand TNF-αwere significantly lower than those of the A group(P<0.05).The levels of LVEF,IL-10,SP,VIP,NK and CD4+/CD8+of the C and D group were significantly higher than those of the B group(P<0.05),while the LVEDD,LVESD,SBP,DBP,NE,AngⅡand TNF-αwere significantly lower than those of the group B(P<0.05).The levels of LVEF,IL-10,SP,VIP,NK and CD4+/CD8+of the D group were significantly higher than those of the C group(P<0.05),while the LVEDD,LVESD,SBP,DBP,NE and AngⅡwere significantly lower than those of the C group(P<0.05).There was no significant difference in the levels of TNF-αbetween the C group and the D group(P>0.05).Conclusion:Salvia miltiorrhiza extract can improve the function of heart and decease the blood pressure of HHD rats,and it suggests that it is likely to play a role by regulating the levels of neuroimmunotic factors.
