摘要
建立高效液相色谱测定金银花中w(绿原酸)的方法,考察φ(乙醇)、m(药材)∶V(提取溶剂)(以下简称"料液比")和提取时间对绿原酸提取率的影响,采用正交实验方法优化提取条件。结果表明,在Agilent HC-C18色谱柱(250mm×4.6mm,5μm),V(乙腈)∶V(磷酸)=15∶85[ρ(磷酸)=0.1g/mL]为流动相,流速为1.0mL/min,检测波长为327nm的色谱条件下,正交实验筛选出绿原酸超声提取的最佳工艺参数为采用料液比35g/mL,提取溶剂φ(乙醇)=60%,超声(200W,40kHz)时间30min。优化的提取工艺操作稳定,结果准确,重复性好。
The HPLC method was established to determine chlorogenic acid in Lonicera japonica and the effects of ethanol concentration,ratio of solid to liquid and ultrasonic extraction time on the contents of chlorogenic acid were studied.The extraction technology of chlorogenic acid was optimized by orthogonal experiment.The results showed that the chromatographic conditions were as follows:Aglient HC-C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid(15∶85),the flow rate was 1.0 mL/min and the detection wavelength was 327 nm.The optimal parameters were determined as follows:the concentration of ethanol was 60%,ratio of solid to liquid was 1∶35 g/mL,ultrasonic extraction(200 W,40 kHz)time was 30 min.The optimized technology is stable,reliable and accurate.
