甲酸提取酿酒酵母中S-腺苷-甲硫氨酸工艺研究

摘要 目的:研究酿酒酵母细胞内S-腺苷-甲硫氨酸(SAM)的提取工艺。方法:以酿酒酵母胞内SAM的提取率为指标,考察6种不同提取剂对SAM的提取效果,确定最佳提取剂;通过比较提取剂浓度、料液比、提取时间、提取温度等因素对SAM回收量的影响,确定最佳提取条件。结果:6种不同提取剂中,甲酸溶液对SAM的提取率最高,达到了85.6%;优选的提取剂浓度为1.2 mol/L、料液比为1:5 g/mL、提取温度为15℃、提取时间为2 h,SAM回收量达12.68mg/g。结论:甲酸提取酿酒酵母胞内SAM是一种提取SAM的新工艺,具有实际应用价值和工业应用前景。 Objective:To study the extraction process for S-adenosyl-L-methionine(SAM)from Saccharomyces cerevisiae.Methods:The optimum extraction solvent was chosen from6different solvents by comparing the extraction rate of intracellular SAM from S.cerevisiae.The effects of solvent concentration,solid-liquid ratio,extraction time and temperature on the yield of SAM were investigated.Results:The extraction rate of SAM was96.6%by formic acid,which was the optimum solvent among the6different solvents.The optimal conditions for maximum SAM yield were obtained at15℃with formic acid of1.2mol/L and solid-liquid ratio of1:5g/mL for2hours.Under such conditions,the extraction yield of SAM was12.68mg/g.Conclusion:It is a novel extraction process for SAM from S.cerevisiae by formic acid solvent,and it has great potential in industrial application.
