海洋细菌ZQM2017产琼胶酶活性测定条件优化 被引量:1

摘要 为研究DNS法测定琼胶酶产生菌所产琼胶酶酶活力的最适条件,确定一组较优的测定琼胶酶活力的条件方法。采用单因素试验法,从缓冲液种类、反应温度和反应p H、反应时间及DNS用量等几个方面研究了海洋细菌ZQM2017产琼胶酶活性测定的适宜条件。研究结果表明:DNS用量,反应时间对酶活都有显著影响,在2 m L反应体系中海洋细菌ZQM2017产琼胶酶酶活测定的最适条件为:酶促反应温度和时间分别为50℃和反应5 min,p H 7. 0,缓冲液种类为磷酸缓冲液,底物浓度6. 5 g/L,DNS用量2. 5 m L。 To study the optimum conditions for the determination of Agar enzyme activity by DNS method,a group of better conditions for the determination of Agar enzyme activity was established.The optimum conditions for the determination of Agar enzyme activity of marine bacteria ZQM2017 were studied from the aspects of buffer type,reaction temperature,reaction pH,reaction time and amount of DNS by single factor test.The results showed that the dosage of DNS and reaction time had significant effects on the enzyme activity.In 2 mL reaction system,the optimum conditions for the determination of Agar enzyme activity of marine bacteria ZQM2017 were as follows:the temperature and time of enzymatic reaction were 50℃ and 5 min at pH 7.0,respectively,the type of buffer solution was phosphoric acid buffer,the substrate concentration was 6.5 g/L ZQM2017 and the dosage of substrate concentration was 2.5 mL.
