摘要
以白及多糖粗提液为对象,考察絮凝剂ZTC-Ⅱ絮凝纯化工艺。在单因素试验的基础上,运用PlackettBurman(PB)试验,选择出具有显著影响的因素。通过满意度函数与响应面相结合的分析方法,建立各影响因素的二次回归方程模型,得到白及多糖的最佳纯化条件:絮凝温度为48℃,絮凝剂ZTC-Ⅱ组分B用量为0. 77 m L/g,组分A、B用量比为0. 34∶1,絮凝时间为1. 5 h,浓缩比为1∶12 g/m L。在此优化工艺下,白及多糖保留率为(92. 32±0. 34)%,脱蛋白率为(51. 43±0. 12)%。
Flocculating purification for crude polysaecharide extracting solution of Bleailla striata by ZTC-Ⅱflocculant was studied.The main influence factors were selected by Plackett-Burman(PB)test on the basis of these single factors.From the results of response surface methodology combined with desirability function,the regression equation of each influencing factor was established.The optimum purification conditions of crude Bleailla striata polysaccharide were as follows:flocculation temperature of 48℃,dosage of component B of 0.77 mL/g,ratio of component A and B of 0.34∶1,flocculation time of 1.5 h,and ratio of mass to volume of 1∶12 g/mL.Under such optimized conditions,the retention rate of polysaccharide was(92.32±0.34)%,and deproteinizing rate was(51.43±0.12)%.
