补肾开窍祛痰方对缺血性脑损伤大鼠海马突触可塑性的影响 被引量:1

摘要 目的探讨补肾开窍祛痰方对缺血性脑损伤大鼠神经损伤的保护作用,阐明其治疗机制。方法 50只SD大鼠随机分为假手术组、模型组、依达拉奉组、补肾开窍祛痰方低和高剂量组,采用改良Longa线栓法制备缺血性脑损伤动物模型,给药7 d后,所有大鼠进行神经功能缺损评分。采用免疫组化法检测海马CA1区神经元突触前膜标记蛋白Synapsin-1和突触后膜标记蛋白PSD-95的表达,Western blot法检测海马突触可塑性功能蛋白Ca MKII、Syn Gap的表达。结果与假手术组比较,模型组大鼠神经损伤明显,海马CA1区Synapsin-1、PSD-95免疫阳性下降(P <0. 01),海马Ca MKII、Syn Gap蛋白表达显著下降(P <0. 01);给予补肾开窍祛痰方干预后,模型大鼠的神经缺损状态有所好转,海马CA1区Synapsin-1、PSD-95免疫阳性显著增强(P <0. 01),Ca MKII、Syn Gap蛋白表达显著上调(P <0. 01)。结论补肾开窍祛痰方能显著改善缺血性脑损伤大鼠的神经损伤情况,其作用可能与调节海马突触可塑性有关。 Objective To explore the protective effect of Bushen-Kaiqiao-Qutan prescription on nerve injury in rats with ischemic brain injury and to clarify its therapeutic mechanism.Methods 50 SD rats were randomly divided into sham,model,edaravone,and low,high-dose of Bushen-Kaiqiao-Qutan prescription group.Animal models of ischemic brain injury were prepared by modified Longa suture method.All rats were scored for neurological deficits after 7 days of administration.Immunohistochemistry was used to detect the expression of presynaptic membrane protein Synapsin-1 and post-synaptic membrane protein PSD-95 in hippocampal CA1 region.The expression of CaMKII and SynGap in hippocampus was detected by Western-blotting.Results Compared with sham group,the rats in model group had significant neurological damage.The Synapsin-1 and PSD-95 immunoreactivity in hippocampal CA1 decreased(P<0.01),and the expression of CaMKII and SynGap protein in the hippocampus was decreased significantly(P<0.01).After intervention of the Bushen-Kaiqiao-Qutan prescription,the neurological deficits of the model rats were improved,Immunostaining of Synapsin-1 and PSD-95 in hippocampal CA1 region was significantly increased(P<0.01),and the expression of CaMKII and SynGap protein was significantly up-regulated(P<0.01).Conclusion Bushen-Kaiqiao-Qutan prescription could significantly improve the neurological damage in rats with ischemic brain injury,which might be related to the regulation of hippocampal synaptic plasticity.
