摘要
采用高温液态水将玉米芯进行预处理,并使用表面活性剂吐温80(Tween 80)辅助纤维素酶进行水解反应,旨在提高生物质催化转化效率。研究结果表明,高温液态水处理可以有效去除半纤维素,使玉米芯表面纤维大量裸露,增加纤维素的可及度;酶催化水解过程中,添加非离子表面活性剂,可以有效屏蔽木质素对纤维素酶的非反应性吸附,改善纤维素与纤维素酶的吸附-解吸附性能;二者相结合,有效的提高了生物质的转化效率。玉米芯经180℃高温液态水处理1 h,酶解过程中Tween 80添加量为0.1 mL/g底物T的条件下,水解液中还原糖浓度(cRS)提高了近三倍。
Corncob was pretreated by hot liquid water(HLW)and then enzymatic hydrolyzed with surfactant assistant Tween 80, aiming at enhancing the biomass conversion efficiency.The results indicated that hemicellulose was effectively removed and lots of cellulose fiber were exposed after HLW pretreatment, thus increased the accessibility for cellulase to hydrolysis cellulose. The presence of nonionic surfactants Tween 80 during enzymatic hydrolysis shielded the nonproductive adsorption of lignin to cellulase and thereby improved the adsorption-desorption property between cellulase and cellulose. After pretreated by HLW at180 ℃ for 1 h, corncob was enzymatic hydrolyzed with Tween 80 dosage of 0.1 mL/g substrate, and the reducing sugar concentration(cRS)in hydrolysate was almost tripled compared with the control experiment.
