柠檬酸对Al^3+致小鼠氧化损伤的治疗作用

摘要 以昆明系雄性小鼠为研究对象,研究柠檬酸对Al^3+致小鼠睾丸组织氧化损伤的影响,以期找出缓解Al^3+所导致的小鼠睾丸组织的氧化损伤。方法:将75只健康雄性昆明系小鼠,随机分为五组,一组为空白对照组,四组为铝攻毒组。分别用PBS缓冲液和60mg/kg·bw氯化铝溶液对空白对照组和铝攻毒组的小鼠进行腹腔注射。两周后将铝染毒组分为柠檬酸低、中、高剂量组和Al^3+对照组;柠檬酸低、中、高剂量组分别注射50mg/kg·bw、90mg/kg·bw、130mg/kg·bwCA,Al^3+对照组注射等量PBS缓冲液。每天均腹腔注射一次,注射量为0.8mL,连续两周,两周后测定小鼠睾丸组织中SOD、GSH-PX酶活性和MDA含量变化。结果:与空白对照组相比,Al^3+对照组SOD活性显著降低,GSH-PX活性却没有显著变化,MDA含量显著增多;与Al^3+对照组相比,柠檬酸中剂量组SOD酶活性显著升高,GSH-PX酶活性没有显著变化,MDA含量显著下降。结论:60mg/kg·bw的氯化铝溶液会引起小鼠睾丸组织的氧化损伤,SOD酶活性显著下降、GSH-PX酶活性没有显著变化、MDA的含量显著变多;中等剂量(90mg/kg·bw)的柠檬酸能缓解Al^3+导致的小鼠睾丸组织的氧化损伤,睾丸组织中SOD活性显著升高、GSH-PX酶活性无变化、MDA含量显著下降。但柠檬酸低(50mg/kg·bw)、高剂量(130mg/kg·bw)对Al^3+所致的氧化损伤没有修复作用。 Aim:In order to relieve the oxidative damage of testis tissue induced by Al^3+,this study investigated the effect of citric acid on the oxidative damage of testis tissue induced by Al^3+ in Kunming(KM)mice.Methods:75 healthy male KM mice were randomly divided into five groups(one blank control group and four Al3+attacked groups).The mice in the blank control group and the Al^3+ attacked groups were injected intraperitoneally with PBS buffer and 60 mg/kg·bw aluminum chloride solution,respectively.After two weeks,the Al^3+ attacked groups were divided into citric acid low,medium,high dose groups,and Al^3+ control group.Mice in the low,medium,and high dose groups were injected with 50 mg/kg·bw,90 mg/kg.bw,130 mg/Kg·bw CA,respectively.Mice in Al^3+ control group were injected with PBS buffer.The activity of SOD,GSH-PX,and the content of MDA in testis were measured after two weeks.Results:Compared with the control group,the SOD activity of mice in Al3+control group were lower significantly than that of the control group,and the activity of GSH-PX were not significantly changed,besides,the content of MDA were significantly increased.Compared with the Al^3+ control group,the activity of SOD,GSH-PX activity of mice in medium dose group did not change significantly,but MDA content were reduced significantly.Conclusion:60 mg/kg·bw of aluminum chloride solution could induce oxidative damage of testis tissue.SOD activity decreased significantly,while GSH-PX activity did not change significantly;moreover,MDA content increased significantly.The medium dose(90 mg/kg·bw)of citric acid had a certain effect on the oxidative damage of testis tissue induced by Al^3+.However,low dose(50 mg/kg·bw)and high dose(130 mg/kg·bw)had no effect on oxidative damage induced by Al3+.
