胰岛结合bFGF生物胶经肾被膜下移植逆转小鼠糖尿病

摘要 目的探讨碱性成纤维细胞生长因子(bFGF)结合重组人胶原蛋白(RHC)与胰岛共移植至糖尿病小鼠肾被膜下逆转同系小鼠糖尿病的效果。方法将分离纯化的Balb/C小鼠胰岛分为三组:(1)Free组(单纯胰岛移植);(2)1 mg/mL RHC+60 ng/mL bFGF组;(3)16 mg/mL RHC+60 ng/mL bFGF组。体外培养6 h后移植入小鼠肾被膜下,移植量为(200±50)IEQ。术后监测小鼠的血糖、体重,并在移植后第30天进行口服糖耐量试验(OGTT),取受体小鼠肾脏移植部位进行组织学评价,观察胰岛在肾被膜下的存活情况。结果移植后30 d,与Free组的血糖[(21.85±0.05)mmol/L]相比,1 mg/mL RHC+60 ng/mL bFGF组[(10.47±2.32) mmol/L,P<0.01]和16 mg/mL RHC+60 ng/mL bFGF组[(8.13±1.73)mmol/L,P<0.01]的血糖水平更低;OGTT结果显示16 mg/mL RHC+60 ng/mL bFGF组小鼠胰岛功能最佳,与正常Balb/C小鼠相似;组织学染色也显示RHC-bFGF组较Free组新生血管更多。结论 bFGF结合RHC与胰岛共移植能达到减少胰岛用量、提高胰岛功能的效果,为胰岛移植的临床应用提供了一种新的思路。 Objective To investigate the reversal effect on the diabetic mice under renal capsules by a biological transplanted glue composing basic fibroblast growth factor (bFGF) and recombinant human collagen (RHC). Methods The isolated and purified Balb/C mice islets were divided into three groups:(1)Free group (only islet transplantation);(2)1 mg/mL RHC+60 ng/mL bFGF group;(3)16 mg/mL RHC+60 ng/mL bFGF group. After 6 h culture in vitro , islets under the equivalent (200±50) IEQ were transplanted into the mice renal capsule. The blood glucose and body weight of the mice were monitored, and the oral glucose tolerance test (OGTT) was performed on the 30th day after transplantation. The kidney transplantation site of the recipient mice was used for histological evaluation to observe the survival of the transplanted islets under the renal capsule. Results Thirty days post-transplantation, compared with the Free group [(21.85±0.05) mmol/L], the 1 mg/mL RHC+60 ng/mL bFGF group [(10.47±2.32) mmol/L, P <0.01] and 16 mg/mL RHC+60 ng/mL bFGF group [(8.13±1.73) mmol/L, P <0.01] had lower blood glucose levels. The OGTT results indicated that function of 16 mg/mL RHC+60 ng/mL bFGF group was optimal and had the same tendency as the normal Balb/C mice. The RHC-bFGF groups had higher levels of neovascularization than Free group from the histological results. Conclusion Islet transplantation combined with bFGF and RHC can reduce the required amount and improve islet function, which will be developed as a new strategy for the clinical application of islet transplantation.
