浙贝母挥发油提取工艺优选及抑菌活性实验研究 被引量:10

Optimized Extraction Process of Volatile Oil from Fritillaria thunbergii Miq. and its Antibacterial Activity

摘要 采用响应面法优化超声-辅助提取浙贝母挥发油工艺,在单因素实验的基础上,考察影响提取工艺的料液比、超声时间和超声温度3个因素,以挥发油得率为响应值,通过三因素三水平实验,确定了其最佳提取工艺,即料液比1︰13g/mL,超声时间25min、超声温度54℃,在该条件下浙贝母挥发油的得率为0.912 7%,与理论值0.919 5%相差较小,说明该方法合理、可行,可用于浙贝母挥发油提取。同时采用纸片法,测试了该挥发油的抑菌活性,结果发现其对大肠埃希菌、金黄色葡萄球菌和肺炎克雷伯氏菌均有一定的抑菌作用,抑菌活性强弱为金黄色葡萄球菌>大肠埃希菌>肺炎克雷伯氏菌。 The extraction process of Fritillaria thunbergii Miq.under ultrasonic assisting was optimized by response surface methodology,and 3 affecting factors(the ratio of material to liquid,ultrasonic time and ultrasonic temperature)of extraction technology were observed based on single factor experiment.The optimized technology was developed through 3-factor and 3-level experiments after determining the volatile oil yield as the response value,and that was:the ratio of solid to liquid,13g/mL,the ultrasonic time,25min,and ultrasonic temperature,54℃.Under this condition,the essential oil yield of Fritillaria thunbergii Miq.was 0.912 7%,which is far smaller different from the theoretical value of 0.919 5%.This result indicated that the method was reasonable and feasible,and could be used for extracting volatile oil from Fritillaria thunbergii Miq..At the same time,the antibacterial activity of the volatile oil was tested by using the paper method.The results indicated that the volatile oil had some bacteriostasis to Escherichia coli,Staphylococcus aureus and Klebsiella pneumoniae,and the antimicrobial activity was Staphylococcus aureus >Escherichia coli >Klebsiella pneumoniae.
