摘要
为筛选与目的基因连锁更加准确的分子标记以及明确分子标记鉴定技术的准确率,同时为后期进行基因精细定位研究提供技术手段,本试验采用F_(2∶3)家系进行F_2单株的TuMV抗性鉴定。结果表明,122个大白菜抗TuMV材料8407和感病材料冠291的F_(2∶3)家系中,抗病的有98个,感病的有24个,经卡方测验,符合3∶1的分离比例,与以往对该群体F_2代TuMV抗性分离比例的研究一致,表明这种鉴定方法可行。本结果可为今后分离群体单株TuMV抗性的准确鉴定提供参考。
In order to screen out molecular markers with more accurate linkage distance and make clear of the identification accuracy of molecular markers,as well as for fine gene mapping in the later period,the F 2∶3 families were used to identify the TuMV resistance of F 2 single plant.The results showed that in 122 Chinese cabbage F 2∶3 families with their resistant parent‘8407’and susceptible parent‘Guan 291’,98 were resistant families and 24 were susceptible families,which was in accordance with the 3∶1 separation ratio by Chi-square test.This was consistent with previous studies in segregation ratio of TuMV resistance in F 2 generation,which indicated that this method was feasible.This study provided a methodological guide for accurate identification of individual isolate on TuMV resistance in the future.
