摘要
利用鼠胚传代培养细胞,制备细胞爬片,并分别运用考马斯亮蓝R250和FITC-鬼笔环肽来染色显示细胞微丝,探索和改进实验方法,以便有效观察细胞微丝的分布和形态结构。结果显示,采用鼠胚传代培养细胞作为实验材料,传代成纤维状细胞铺展良好,经固定、通透并染色后,可在普通光学显微镜和荧光显微镜下显示微丝束在细胞中的分布,既便于实验观察,也有助于学生对微丝骨架作用的理解和掌握。
In order to improve experimental methods of the cell microfilament observation experiments,mouse embryonic subcultured cells were used as experimental materials,and well passaged on the glass coverslips.After the cells were fixed and permeabilized,cell microfilaments were stained and displayed using Coomass R250 and FITC-Phalloidin respectively.The results showed that under the bright microscope and fluorescence microscope,the cell microfilaments can be displayed more clearly and effectively.It is easy for experimental observation and helps students understand the role of the microfilament cytoskeleton.
