摘要
该试验选取一株分离自辽河油田原油的菌株ZY-1,分别补加培养基碳源(木糖、淀粉、葡萄糖),于60℃静置培养7 d,通过测定发酵液的菌体密度、表面张力、乳化活性以及石蜡的残留量,考察菌株ZY-1利用上述碳源的生长以及发酵液表面活性情况。结果表明,菌株ZY-1可利用木糖生长,并代谢产生物表面活性剂。补加木糖能明显促进细胞生长,减慢对石蜡的利用,使发酵液表面张力降至49.5 m N/m,发酵液的乳化活性提高至61.3%。补加葡萄糖或淀粉虽不能明显促进细胞生长,但能明显减少石蜡消耗,都能将发酵液表面活性提高至63.3%。木糖、葡萄糖和淀粉都能减少石蜡的消耗,木糖效果更明显。
Strain ZY-1 isolated from Liaohe crude oil for enhancing oil recovery was cultivated at 60℃for 7 d in the media with xylose,starch and glucose as carbon sources.The growth of strain ZY-1 and surface activity of strain fermentation broth were evaluated by measuring the cell density,surface tension,emulsifying activity and the residues of paraffin.The results showed that the strain ZY-1 could utilize the tested carbon sources for the growth of cells and product biosurfactants by metabolism.The mixed carbon source of paraffin and xylose could obviously improve the growth of cells,slow down the utilization of paraffin,reduce surface tension of fermentation broth to 49.5 mN/m,and increase the emulsification activity of the fermentation broth to 61.3%.Xylose could significantly reduce the consumption of paraffin.The mixed carbon source of paraffin and glucose or starch could not obviously promote cell growth,but obviously reduce the paraffin consumption and enhance the surface activity of fermentation broth to 63.3%.Xylose,glucose and starch could reduce the consumption of paraffin,xylose was more effective.
