摘要
目的研究麻杏石甘合剂对LPS联合香烟熏蒸所诱导的COPD模型大鼠肺部功能的干预作用及作用机制。方法将SD大鼠于实验第1、14天通过气道滴注生理盐水,并于30 d后检测以此作为空白组;另取SD大鼠于实验第1、14天通过气道滴注脂多糖(LPS)200μg/只,并联合香烟烟雾诱导每天2 h,共30 d,建立模型组。其他各实验组在造模的同时分别给予相应受试药物,通过观察各组中大鼠肺部呼吸功能、血清中TNF-α、IL-6含量及肺组织形态学的变化,探讨麻杏石甘合剂对COPD模型大鼠肺功能的干预作用。结果通过与空白组比较,模型组大鼠呼吸功能参数值FVC、FEV0.3、PEF明显下降(P<0.01);大鼠血清中IL-6、TNF-α含量明显升高(P<0.01);通过与模型组比较,麻杏石甘合剂高中剂量组与阳性对照药氨茶碱给药组使大鼠肺部呼吸功能参数值FVC、FEV0.3、PEF得到明显改善(P<0.01或P<0.05);血清中IL-6、TNF-α含量降低(P<0.01或P<0.05);肺组织病变程度减轻。结论通过给予麻杏石甘合剂可通过改善肺组织病变,提高肺部呼吸功能,降低炎性因子的产生来实现其对慢性阻塞性肺疾病的干预及预防作用。
Objective To study the intervention effect and mechanism of Maxing Shigan mixture on lung function of rats with COPD induced by LPS combined with cigarette fumigation.Methods SD rats were instilled with saline on the 1st and 14th day of the experiment,and were detected after 30 d,which were set as the blank group.SD rats instilled with lipopolysaccharide(LPS)200μg/rat by airway on the 1st and 14th day of the experiment,combined with cigarette smoke induction with 2 hours a day for 30 d.And model group was established.The other experimental groups were given the corresponding test drugs while modeling.The lung respiratory function,serum TNF-α,IL-6 content and lung tissue morphology change were observed in each group to explore the intervention role of Maxing Shigan mixture on pulmonary function in rats with COPD model.Results Compared with those of the blank group,the respiratory function parameters FVC,FEV0.3,and PEF of the model group were significantly decreased(P<0.01),and the serum levels of IL-6 and TNF-αwere significantly increased in the rats of the model group(P<0.01).Compared with the model group,the high-dose group of Maxing Shigan mixture and the positive control drug aminophylline group improved the respiratory function parameters FVC,FEV0.3 and PEF of the rats(P<0.01 or P<0.05),serum IL-6 and TNF-αlevels were decreased(P<0.01 or P<0.05).The extent of lung tissue lesions was reduced.Conclusion By administering Maxing Shigan mixture,it can improve the lung tissue damage,improve lung respiratory function and reduce the production of inflammatory factors to achieve its intervention and prevention of chronic obstructive pulmonary disease.
