摘要
目的:建立柱前衍生高效液相色谱(PMP-HPLC)法,测定太子参中葡萄糖的含量。方法:高温强酸水解将太子参多糖水解为单糖,1-苯基-3-甲基-5-吡唑啉酮(PMP)将其衍生化;采用HPLC法,Hypersi L BDS C18色谱柱,15%(v/v)乙腈+50 mmol/L磷酸缓冲液(KH_2PO_4-NaOH,pH 6.9)和40%(v/v)乙腈+50 mmol/L磷酸缓冲液(KH_2PO_4-NaOH,pH 6.9)为流动相,梯度洗脱,流速为1.0 mL/min,在250 nm波长检测太子参中的葡萄糖单糖。结果:葡萄糖在0.52~15.62 mg/mL线性范围内呈良好的线性关系(r=0.9990),回归方程为y=41211.22x-857.23,平均加样回收率分别为97.96%、99.45%、109.37%。结论:该方法重现性好、准确度高,为太子参药材质量标准的完善奠定基础。
Objetive To establish a Pre-column Derivatization High Performance Liquid Chromatography(PMP-HPLC)method for the determination of glucose in Pseudostellaria heterophylla.Methods Polysaccharides of Pseudostellaria heterophylla was hydrolyzed into monosaccharides by high-temperature strong acid hydrolysis,and 15%(v/v)acetonitrile+50 mmol/L phosphate buffer(KH2PO4-NaOH,pH 6.9)and 40%(v/v)acetonitrile+50 mmol/L phosphate buffer were used by HPLC.Results Glucose monosaccharide in Pseudostellaria heterophylla was determined by gradient elution at a flow rate of 1.0 mL/min and a linear range of 0.52~15.62 mg/mL(r=0.9990).The regression equation was y=41211.22 x-857.23.The average recovery was 97.96%,99.45%and 109.37%respectively.Conclusion the method has good reproducibility and high accuracy,and lays the foundation for improving the quality standard of Radix pseudo ginseng.
