摘要
目的:研究PI3K/AKT信号通路在TNF-α诱导上皮间质转化(EMT)发生时促口腔鳞癌侵袭转移中的作用。方法:采用免疫蛋白印迹实验(western blot)方法、实时荧光定量RT-PCR,分别从蛋白质、mRNA水平检测炎症微环境中PI3K/AKT信号通路抑制剂作用前后关键因子表达变化。结果:TNF-α作用后,PI3K/AKT信号通路中关键因子AKT蛋白表达升高,E-钙黏蛋白mRNA表达降低,而加入相应抑制剂LY294002后,AKT的蛋白表达被抑制,E-钙黏蛋白较前者表达升高。结论:抑制PI3K/AKT信号通路可降低TNF-α诱导EMT发生促口腔鳞癌细胞侵袭转移的作用。
Objective: To observe the role of PI3K/AKT signaling pathway in promoting the invasion and metastasis of oral squamous cell carcinoma induced by TNF-α in patients with epithelial-mesenchymal transition (EMT). Methods :Western blot and real-time PCR were performed to detect the expression of key factors in inflammatory microenvironment before and after treatment with PI3K/AKT signal pathway inhibitor. Results :After TNF-α treatment,the expression of key factor AKT protein in PI3K/AKT signaling pathway was increased and E-cadherin mRNA expression was significantly decreased.The protein expression of PI3K/AKT was significantly inhibited after addition of corresponding inhibitor LY294002,and E-cadherin expression was significantly higher than that of the former. Conclusion :Inhibition of PI3K/AKT signaling pathway can weaken the role of EMT induced by TNF-α in promoting the invasion and metastasis of oral squamous carcinoma cells.
