摘要
目的建立绞股蓝药材中绞股蓝皂苷XLIX含量的高效液相色谱-蒸发光散射(HPLC-ELSD)法。方法色谱柱为Dubhe C18柱(250 mm×4. 6 mm,5μm),流动相为乙腈-水(梯度洗脱),流速为1 m L/min,检测器为蒸发光散射检测器,流速为2. 7 L/min,雾化温度为99. 7℃。结果绞股蓝皂苷XLIX进样量在2. 061 5~12. 936 0μg范围内线性关系良好,平均回收率为100. 12%,RSD为1. 17%(n=9)。结论该法简单快速,专属性强,重复性良好,可用于绞股蓝皂苷XLIX的质量控制。
Objective To establish an HPLC-ELSD method for the content determination of gypenoside XLIX in Gynostemma pentaphyllum.Methods The chromatographic column was Dubhe C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-water(gradient elution),the flow rate was 1 mL/min.ELSD detector was adopted with flow rate of 2.7 L/min and the atomization temperature was 99.7℃.Results The gypenoside XLIX showed a good relationship within the range of 2.061 5-12.936 0μg,the average recovery rate was 100.12%,RSD was 1.17%(n=9).Conclusion The method is simple,rapid,specific and reproducible,which can be used for the quality control of gypenoside XLIX in Gynostemma pentaphyllum.
