摘要
目的探讨广西某铝矿区老年人群血清中铝元素含量与β-分泌酶1(BACE1)、早老素1(PS1)、早老素2(PS2)、分拣蛋白相关受体(SORL1)和缺氧诱导因子(HIF1)的mRNA表达的相关性。方法根据简易智力状态检测(MMSE)得分,将受试者分为非矿区对照组和非矿区病例组,矿区对照组和矿区病例组。采用电感藕合等离子体质谱法检测各组血液中铝元素含量及采用实时荧光定量PCR法检测血液中BACE1、PS1、PS2、SORL1和HIF1的mRNA表达量,并分析铝元素含量与BACE1、PS1、PS2、SORL1和HIF1的mRNA表达相关性。结果矿区病例组老年人血清中铝元素含量显著增高(P <0.01),血清中BACE1、PS1、PS2的mRNA表达量升高(P <0.01),SORL1的mRNA表达量降低(P<0.01)。对矿区与非矿区受试老年人血清中铝元素与BACE1、PS1、PS2、SORL1基因的mRNA的相关性分析,相关系数分别为0.489、0.485、0.476、-0.415,差异有统计学意义(P <0.05),与HIF1基因的mRNA的相关系数为0.342,差异无统计学意义(P>0.05)。提示铝元素含量与BACE1、PS1、PS2的mRNA表达量呈正相关,与SORL1的mRNA表达量呈负相关。结论老年性痴呆症(AD)的前期症状——认知障碍的发生可能是因为浅表铝暴露致使人体内铝元素含量升高而导致了AD相关遗传危险因子BACE1、PS1、PS2的mRNA表达量升高,SORL1的mRNA表达量降低而引起的。
Objective To investigate the correlation between serum aluminum content and mRNA expression ofβ-site APP cleaving enzyme 1(BACE1),presenilin-1(PS1),presenilin-2(PS2),sortilin-related receptor 1(SORL1)and hypoxia-inducible factor 1(HIF1)in elderly population from an aluminum mining area in Guangxi.Methods According to mini-mental state examination(MMSE)scores,the elderly people were divided into non-mining-area control group and non-mining-area case group,mining-area control group and mining-area case group.Inductively coupled plasma mass spectrometry was used to detect the content of blood aluminum for each group and real-time fluorescence quantitative PCR was used to measure the mRNA expression of BACE1,PS1,PS2,SORL1 and HIF1 in blood,and the correlation between the content of aluminum and the mRNA expression of BACE1,PS1,PS2,SORL1 and HIF1 was analyzed.Results The content of aged serum aluminum in mining-area case group was significantly increased(P<0.01),and the mRNA expressions of serum BACE1,PS1 and PS2 were increased(P<0.01),and the mRNA expression of SORL1 was decreased(P<0.01).The correlation analysis between elderly serum aluminum and mRNA expression of BACE1,PS1,PS2 and SORL1 from mining and non-mining areas showed that the correlation coefficients were 0.489,0.485,0.476 and-0.415,respectively,with significant difference(P<0.05),and the correlation coefficient with HIF1 was 0.342,with no statistical difference(P >0.05).It indicated that the content of aluminum was positively correlated with the relative mRNA expression of BACE1,PS1 and PS2,and negatively correlated with the mRNA expression of SORL1.Conclusion The early symptom of Alzheimer’s disease(AD)-cognitive impairment may be caused by the increase of aluminum content in human body caused by superficial aluminum exposure,which leads to the increase of mRNA expression of AD related genetic risk factors BACE1,PS1 and PS2,and the decrease of mRNA expression of SORL1.
