淫羊藿女贞子配伍提高哮喘大鼠对地塞米松作用的敏感度研究 被引量:4

摘要 目的探讨中药淫羊藿女贞子合用的煎剂及其提取物在地塞米松干预大鼠哮喘的模型中,对于糖皮质激素受体(GR)作用途径的影响,以评价中药提高哮喘大鼠对糖皮质激素作用的敏感度。方法 SPF级雄性SD大鼠卵蛋白诱发哮喘造模,在地塞米松治疗的同时给予淫羊藿女贞子合用的煎剂或提取物。采用免疫组化、Western blot法及实时荧光PCR技术检测肺组织GRα、GRβ、HSP90的蛋白及m RNA的表达。结果与正常组相比,模型组肺组织中GRα与HSP90蛋白和m RNA的表达、GRβm RNA的表达、GRα/GRβ,GRα/HSP90在m RNA水平和GRβ/HSP90在蛋白水平有统计学意义(P<0.05或P<0.01)。与模型组比较,地塞米松组GRα表达明显下调(P<0.05);淫羊藿女贞子煎液及提取物单用及合用地塞米松组均能明显上调肺组织中GRα和HSP90的表达、GRα/GRβ和GRα/HSP90在m RNA水平的比值(P<0.05或P<0.01),抑制GRβm RNA表达以及GRβ/HSP90(P均<0.01)。结论淫羊藿女贞子煎剂及提取物均能通过改善GR亚型与HSP90的表达,调整GR作用途径而提高哮喘大鼠对地塞米松作用的敏感度。 Objective To research the influences of the decoction and their extracts of Herba Epimedii and Fructus Ligustri Lucidi on glucocorticoid receptor(GR)in the asthmatic rats with dexamethasone intervention.Methods Ovalbumin sensitized rat asthmatic model.With the oral administration of the decoction and their extracts of Herba Epimedii and Fructus Ligustri Lucidi accompanied by the intraperitoneal injection of dexamethasone,the protein and mRNA expressions of GRα,GRβand HSP90 in lung tissue were observed.Results Compared with the normal group,the protein and mRNA expressions of GRαand HSP90,the mRNA expression of GRβ,GRα/GRβ,the ratio of GRαmRNA to HSP90 mRNA,and the ratio of GRβprotein to HSP90 protein in the asthmatic rats were significantly different(P<0.05,P<0.01).Compared with model group,the protein and mRNA expressions of GRαin dexamethasone group significantly increased(P<0.05).The protein and mRNA expressions of HSP90 and GRα,the mRNA expression of GRβ,GRα/GRβand GRα/HSP90 in mRNA level,and GRβ/HSP90 in protein and mRNA levels were significant differences(P<0.05,P<0.01).Conclusion The decoction and their extracts of Herba Epimedii and Fructus Ligustri Lucidi could enhance the sensitivity of the asthmatic rats to dexamethasone through improving subtypes of GR and HSP90.
