尘肺患者外周血miR-21表达与细胞因子分泌、胶原代谢及氧化应激的相关性 被引量:6

摘要 目的分析尘肺患者外周血miR-21表达与细胞因子分泌、胶原代谢、氧化应激的相关性。方法选择2015年3月-2018年2月四川省攀枝花市第二人民医院职业病科诊治尘肺患者43例作为尘肺组,同期在医院进行健康体检的志愿者60例作为对照组。测定2组外周血miR-21的表达量以及血清细胞因子、胶原代谢指标、氧化应激指标的含量。结果尘肺组患者外周血miR-21表达量以及血清转化生长因子-β_1(TGF-β_1)、肿瘤坏死因子-α(TNF-α)、结缔组织生长因子(CTGF)、基质金属蛋白酶(MMP-9)、纤溶酶原激活物(uPA)、胶原蛋白-I(Col-I)、胶原蛋白-Ⅲ(Col-Ⅲ)、8-异前列腺素F2α(8-iso-PGF2α)、一氧化氮(NO)含量均明显高于对照组(t/P=19.837/0.000、16.622/0.000、23.490/0. 000、18. 328/0. 000、26. 239/0. 000、21. 257/0. 000、10. 211/0. 000、13. 912/0. 000、11. 634/0.000、9. 646/0.000),血清总抗氧化力(T-AOC)、超氧化物歧化酶(SOD)含量均明显低于对照组(t/P=18. 790/0.000、16.078/0.000);外周血miR-21表达量与血清TGF-β_1、TNF-α、CTGF、MMP-9、uPA、Col-I、Col-Ⅲ、8-iso-PGF2α、NO的含量呈正相关(r/P=0. 652/0.004、0.474/0.024、0.512/0.018、0.423/0. 029、0. 559/0.020、0. 609/0. 009、0. 541/0.014、0.629/0.008、0.635/0.006),与血清T-AOC、SOD含量呈负相关(r/P=-0. 577/0.013、-0. 519/). 019)。结论尘肺患者外周血miR-21的高表达与病程中细胞因子异常分泌、胶原代谢紊乱、氧化应激激活有关。 Objective To analyze the correlation between the expression of microRNA-21( miR-21) in peripheral blood of pneumoconiosis patients and cytokine secretion, collagen metabolism and oxidative stress. Methods From March2015 to February 2018, 43 patients with pneumoconiosis in the occupational disease department of the Second People’s Hospital of Panzhihua City, Sichuan Province, were selected as the pneumoconiosis group, and 60 volunteers who underwent health examination in the hospital during the same period were selected as the control group. The expression of miR-21 in peripheral blood and the levels of serum cytokines, collagen metabolism and oxidative stress were measured. Results The expression of miR-21 in peripheral blood of patients with pneumoconiosis and serum transforming growth factor-β1(TGF-β1), tumor necrosis factor alpha(TNF-α), connective tissue growth factor(CTGF), matrix metalloproteinase(MMP-9), plasminogen activator(uPA), collagen I(Col-I), collagen III(Col-III), 8-isoprostaglandin-F2α(8-iso-PGF2α), and nitric oxide(NO) were significantly higher than the control group(t/P = 19. 837/0. 000, t/P = 16. 622/0. 000, t/P = 23. 490/0. 000, t/P =18.328/0.000, t/P= 26.239/0. 000,t/P= 21. 257/0. 000,t/P = 10. 211/0. 000,t/P = 13. 912/0. 000,t/P = 11.634/0.000,t/P =9. 646/0. 000), serum total antioxidant capacity(T-AOC), The content of superoxide dismutase(SOD) was significantly lower than that of the control group(t/P = 18. 790/0. 000, t/P = 16. 078/0. 000);the expression of miR-21 in peripheral blood and serum TGF-β1, TNF-α, CTGF, MMP-9, uPA, Col-I there was a positive correlation between the contents of Col-Ⅲ and 8-iso-PGF2α and NO(r/P = 0. 652/0. 004, r/P = 0. 474/0. 024, r/P = 0. 512/0. 018, r/P = 0. 423/0.029,r/P= 0.559/0.020,r/P = 0.609/0.009, r/P =0.541/0.014, r/P = 0.629/0.008,r/P=0.635/0.006), negatively correlated with serum T-AOC and SOD content(r/P =-0. 577/0. 013, r/P =-0. 519/0. 019). Conclusion The high expression of miR-21 in peripheral blood of pneumoconiosis patients is related to abnormal secretion of cytokines, disorder of collagen metabolism and activation of oxidative stress in the course of pneumoconiosis.
