一种制备高存活性肠膜明串珠菌冻干制剂的方法 被引量:1

A Method to Prepare Lyophilized Freeze-dried Preparation of Leuconostoc mesenteroides Powder with High Bacterial Cell Survivality

摘要 为制备高活性冻干粉,通过响应面法对BD1710菌株的培养条件进行优化,结果显示:当培养时间为29.5 h,培养温度为29.5℃,初始p H值为7.3,蔗糖浓度为16.8%时,菌株BD1710冻干存活率可达92.44%,与理论值基本一致,并显著优于传统工艺的冻干保护效果(76.81%)。推测可能是BD1710合成的胞外多糖以及培养基中的天然冻干保护剂为冻干过程中的微生物细胞提供了主要的细胞保护。因此,发酵后直接冻干可作为制备高活性肠膜明串珠菌制剂的一种简易手段。 To prepare lyophilized powder of Leuconostoc mesenteroides BD1710 with high bacterial cell survivality,the cultivation conditions were optimized by the response surface methodology.A practice survivality of 92.44% of the bacterial cells of the strain BD1710 was obtained by directly lyophilizing the bacterial culture obtained under the optimized cultivation conditions,which was composed of tomato juice supplemented with 168 g/L sucrose(with the initial pH value of the broth adjusted to 7.3),and cultivation at 29.5℃for 29.5 h,which was in general agreement with the predicted value and significantly higher than of the survivality(76.81%)of the bacterial cells obtained by cultivating the strain BD1710 in M17 after the cells were lyophilized at the same condition.It is postulated the exopolysaccharides synthesized by the strain BD1710 and natural protective agent in the medium provided the predominant cryptoprotection to the bacterial cells during the freezen-drying process.Therefore,the directly freezen-drying the bacterial cells obtained under cultivation conditions provided a simplified method to prepare lyophilized Leuconostoc mesenteroides powder with high bacterial cell survivality.
