摘要
目的:观察氟化钠对大鼠心肌caspase-3、caspase-8 mRNA表达的影响。方法:通过大鼠自由饮用含不同浓度氟化钠的去离子水的方法制备慢性氟中毒模型,观察各组大鼠的氟斑牙形成情况,RT-PCR法比较各组大鼠caspase-3、caspase-8 mRNA表达情况。结果:4个月后,饮用含氟水的大鼠均出现了不同程度的氟斑牙,随饮水中氟浓度的增加而加重,并且随着饮用时间的延长,大鼠氟斑牙情况加重;与对照组相比,4个月、7个月和10个月后,饮用含NaF水的大鼠心肌caspase-3和caspase-8基因表达增加,且随时间的延长,两者的表达量增加(P<0.05)。结论:慢性氟中毒可能通过诱导大鼠心肌caspase-3、caspase-8基因表达增加,促进心肌细胞凋亡。
Objective:To observe the effects of sodium fluoride on the expression of caspase-3 and caspase-8 mRNA in rat myocardium.Methods:Chronic fluorosis model was prepared by freely drinking deionized water containing different concentrations of sodium fluoride in rats.The formation of dental fluorosis in each group of rats was observed.And mRNA levels of caspase-3 and caspase-8 of different groups in rats were determined by RT-PCR.Results:After drinking fluoride-containing water 4 months,different degrees of dental fluorosis of rats were showed,which was aggravated with the increase of fluoride concentration in drinking water.In addition,after 7 months and 10 months of drinking,the dental fluorosis in rats was more serious.Compared with the control group,after 4 months,7 months and 10 months,the expressions of caspase-3 and caspase-8 genes in the myocardium of rats drinking NaF water increased,and the expressions of caspase-3 and caspase-8 increased with the passage of time(P<0.05).Conclusion:Chronic fluorosis may promote the apoptosis of myocardial cells by inducing the expression of caspase-3 and caspase-8 in rat myocardium.
