嗜酸乳杆菌IMAU30067增殖培养基的研究 被引量:9

摘要 嗜酸乳杆菌IMAU30067是分离自新疆传统酸马奶中的一株具有潜在益生特性的乳酸菌。本研究针对嗜酸乳杆菌IMAU30067的营养需求,通过增殖培养基优化,获得其高密度培养条件。采用单因素试验、正交试验以及响应面法对嗜酸乳杆菌IMAU30067的培养基成分以及静态培养条件进行优化。通过优化得到嗜酸乳杆菌IMAU30067的最佳增殖培养基为:葡萄糖30.00 g/L、麦芽糖30.00 g/L、海藻糖20.00 g/L、鱼蛋白胨30.00 g/L、大豆蛋白胨10.00 g/L、柠檬酸钠2.29 g/L、乙酸钠5.74 g/L、K_2HPO_42.29 g/L、MgSO_4·7H_2O 0.80 g/L、马铃薯提取物6.00 g/L、组氨酸0.10 g/L。最佳培养条件为:接种量1×10~6CFU/mL、初始pH值6.5,于37℃恒pH5.0厌氧培养。最后利用5L发酵罐进行小试发酵试验,优化后IMAU30067的活菌数达3.72×10~9CFU/mL。通过对IMAU30067增殖培养基及培养条件的优化,IMAU30067活菌数较MRS基础培养基提高了82.6倍,为其高密度培养奠定了基础。 Lactobacillus acidophilus IMAU30067 which was isolated from traditional koumiss of Xinjiang has potentially probiotic properties.In this study,the nutrient requirement of Lactobacillus acidophilus IMAU30067 was optimized by the culture medium to obtain high density of cul-ture conditions.The tests use single factor experiment,orthogonal experiment and response surface methods to optimize the enrichment me-dium composition and static culture conditions of Lactobacillus acidophilus IMAU30067.The optimal culture medium for Lactobacillus acidophilus IMAU30067 was glucose 30.00 g/L,maltose 30.00 g/L,Trehalose 20.00 g/L,fish peptone 30.00 g/L,soy peptone 10.00 g/L,sodium citrate 2.29 g/L,sodium acetate 5.74 g/L,K2HP04 2.29 g/L,MgS04·7H20 0.80 g/L,potato extract 6.00 g/L,histidine 0.10 g/L.the optimized re-sults were as follows:The inoculum size is1×10^6 CFU/mL,The initial pH value is 6.5,The constant temperature is 37℃,The constant pH5.0.Using 5 L fermentation tank for test of small test fermentation,The number of the living bacteria cultured by the optimal culture pro-cess can up to 3.72×10^9 CFU/mL.By studying the optimization of the enrichment medium of IMAU30067 and its culture conditions,the viable count of IMAU30067 increases of 82.6 times comparing to the MRS culture,and this result lays a great foundation for the high-densi-ty culture of IMAU30067.
