摘要
目的:观察电针"太冲""百会"对自发性高血压大鼠(SHR)肾RAS系统中舒血管活性因子的影响,探讨其对高血压肾损害的保护机制。方法:24只雄性SHR大鼠随机分为电针组和模型组,12只雄性Wistar-Kyoto(WKY)大鼠作为空白组,电针组电针两极分别连接右侧"太冲穴"和"百会穴"(1 m A,2Hz/15 Hz,20 min)。观察各组血压变化,检测血清中尿酸(UA)和尿素氮(BUN)的含量、肾组织ACE2、Ang(1-7)、AngⅡ的表达,计算Ang(1-7)/AngⅡ的比值; HE、Masson染色和电子透射显微镜观察肾脏结构形态变化。结果:电针组和模型组相比,收缩压、血清中UA、BUN的含量、AngⅡ表达均有所下降(P <0. 05);肾组织ACE2、Ang(1-7)的表达水平、Ang(1-7)/AngⅡ比值提高(P <0. 05)。HE、Masson染色以及电镜观察结果均显示电针组较模型组肾损害现象减轻。结论:电针"太冲""百会"穴能降低血压,减轻高血压对肾的损害。
Objective: To observe the effect of electro-needling Taichong( LR3) and Baihui( GV20) on vasoactive factors of RASS in spontaneous hypertensive rats( SHRs),and to explore its underlying protective mechanism of renal injury in hypertension. Methods: 24 male SHRs were randomized into the model group and the electro-acupuncture( EA) group,another 12 Wistar-Kyoto( WKY) rats were taken as the blank control.Points chosen in the EA group were right LR3 and right GV20( 1 mA,2 Hz/15 Hz,20 min). After 30 days of the treatment,the changes of Bp were observed,serum levels of blood uric acid( UA) and blood urea nitaogen( BUN) were detected,renal expressions of ACE2,Ang( 1-7),and AngⅡ were determined,as well as Ang( 1-7)/AngⅡ was calculated. Pathological changes of renal tissues were observed under HE staning,Masson staining and electron microscope. Results: In EA group,systolic pressure( SP) and the serum contents of AngⅡ,UA and BUN were decreased compared to those in the model group( P < 0. 05). The expressions of ACE2,Ang( 1-7),and Ang( 1-7)/AngII were increased in the kidney in the EA group compared to those in the model group( P < 0. 05). The results of HE staining,Masson staining and electron microscope showed that the kidney damage was less in the EA group than that in the model group. Conclusion: Electro-needling LR3 and GV20 can effectively reduce SP and relieve the damage of high blood pressure to kidney.
