成年火龙果植株茎段愈伤组织诱导、增殖及分化研究 被引量:8

摘要 【目的】为建立和优化火龙果的再生体系。【方法】以成年植株茎段作为外植体,研究了不同表面灭菌方法、不同取样时间及取样部位对外植体污染、褐化的影响,以及不同植物生长调节剂浓度及配比、培养条件等对茎段愈伤组织诱导、增殖及分化的影响。【结果】适宜幼嫩茎段表面灭菌的方式为75%酒精消毒30 s+0.1%HgCl_2消毒6~8 min,适宜成熟茎段表面灭菌的方式为75%酒精消毒30 s+0.1%HgCl_2消毒12~15 min;以春季萌发的10~20 cm枝条诱导愈伤组织效果较好,最适愈伤组织诱导的培养基为MS+TDZ 0.4 mg/L+KT 0.8 mg/L,暗培养7 d,再转入光照培养,愈伤组织为黄绿色、致密状,外表有颗粒状突起。愈伤组织增殖培养基以MS+TDZ 0.4 mg/L+KT(或ZT) 0.8 mg/L效果较好,增殖系数在8倍以上。在愈伤组织分化过程中,添加20%~30%的椰子水对不定芽的形成具有一定促进作用,但分化率较低。【结论】以成年火龙果植株茎段建立的再生体系,愈伤组织分化率总体表现较低,最高分化率仅13%,因此还有待进一步加强和完善。 【Objective】 The persent paper was conducted to establish and optimize the regeneration system of Hylocereus undatus.【Method】 The effects of different surface sterilizing methods, sampling date, sampled parts, concentration and ratio of growth regulators, and culture conditions on contamination rate, browning rate, callus induction, proliferation and differentiation of stem segments collected from adult Hylocereus undatus plants were studied.【Result】 The optimal surface sterilizing method of young and adult stem segments was 75 % alcohol + 0.1 % HgCl2 for 6-8 min and 75 % alcohol + 0.1 % HgCl2 for 12-15 min respectively. The callus induction rate of new germinated stem segments with 10-20 cm length in Spring was better. The optimal medium for callus induction of stem segments was MS + 0.4 mg/L TDZ + 0.8 mg/L KT. The inducted callus after 7 days dark culture and continuous light culture was a yellow green compact tissue with exsert granulates. The optimal medium for callus proliferation was MS + 0.4 mg/L TDZ + 0.8 mg/L KT(or ZT) and the proliferation coefficient was above 8 times. Adding 20 %-30 % coconut water in the differentiation media had a certain promotion effect on adventitious buds formation of calli but the differentiation rate still was lower.【Conclusion】The established regeneration system of stem segments from adult Hylocereus undatus plants with only 13 % differentiation rate of adventitious buds should be further improved.
