常用技术检测河北鼠疫腐败材料的效果分析 被引量:3

Detection effect of the commonly used technology for the corrupt materials of rodents infected with plague in Hebei Province

摘要 目的评价不同检测技术检测河北鼠疫监测工作中腐败材料的应用效果。方法运用细菌培养、胶体金免疫试纸条(GICA)、反向血凝试验(RIHA)、酶联免疫吸附试验(ELISA)以及聚合酶链式反应(PCR)共5种检测技术对255份鼠脏器材料(肝脾混合物)新鲜时和腐败后分别进行检测,记录实验数据,进行统计分析。结果细菌培养对新鲜标本检测敏感性为100%,特异性为100%;材料腐败后,鼠疫菌培养检出受限,敏感性仅为16. 7%,经卡方检验,有统计学意义。标本腐败对胶体金免疫层析、反相血凝试验、酶联免疫吸附试验和聚合酶链式反应检测结果影响较小,经卡方检验,P值均远远大于0. 05,无统计学意义。结论细菌培养受材料腐败的影响很大,但它是唯一能得到纯菌的方法;抗原检测方法中,胶体金免疫层析技术操作最简单,敏感性和特异性受材料腐败的影响较小,可列为监测中筛选阳性标本的首选方法;聚合酶链式反应是基于分子水平的检测,受材料腐败的影响很小。在实际监测工作中,要联合运用细菌培养,抗原检测和基因检测,以提高鼠疫菌的检出率。 Objective To evaluate the detection effect of commonly used techniques for the corrupt materials of rodents infected with plague.Methods Two hundred and fifty-five specimens of corrupted rat visceral organs(liver and spleen)were served as test material,and five techniques of bacteria culture,colloidal gold immune strip(GICA),RIHA,ELISA,and PCR were adopted for the tests.The experimental data were recorded and analyzed statistically.Results Bacterial culture test held the sensitivity of 16.7%,while its specificity reached 100%.Chisquare test showed that there was statistical significance.The results of GICA,RIHA,ELISA and PCR were slightly affected by sample spoilage.Chi-square test showed that the P value was much higher than 0.05,and there was no statistical significance.Conclusion Bacteria culture was highly affected by the material corruption,the colloidal gold immune chromatography technology was the most simple operation with high sensitivity and specificity,and polymerase chain reaction was little affected by the material corruption.In practical monitoring,it is suggested that techniques of bacterial culture,antigen detecting,and gene test should be used jointly.
