摘要
目的探讨缓脾1号方对链脲佐菌素(STZ)诱导糖尿病大鼠糖脂代谢的影响,并探讨其可能的作用机制。方法 60只雄性SD大鼠随机分为空白组、模型组、吡格列酮组及缓脾1号低剂量组、中剂量组、高剂量组,每组10只。除空白组外其余各组采用高糖高脂饲料联合腹腔注射STZ建立糖尿病大鼠模型,吡格列酮组给予吡格列酮17.5 mg/(kg·d),缓脾1号低、中、高剂量组分别给予缓脾1号0.3、0.7、1.4 g/(kg·d)灌胃。空白组与模型组给予等量生理盐水(1 mL/100 g)灌胃,1次/d。各组均连续给药4周。分别于给药后检测空腹血糖(FPG)、口服葡萄糖耐量实验(OGTT)、血清游离脂肪酸(FFA)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL),ELISA法检测肝组织中丙二醛(MDA)、总超氧化物歧化酶(SOD)活性,HE染色法观察肝组织形态学变化。结果与模型组相比,缓脾1号高剂量组OGTT 30 min、60 min、120 min时点血糖值均明显降低(P<0.01,P<0.05);缓脾1号高剂量组、中剂量组血清FFA、TC、TG、LDL及缓脾1号低剂量组血清TC、TG、LDL明显降低(P<0.01,P<0.05);缓脾1号各剂量组肝组织SOD水平升高,MDA水平降低(P<0.01,P<0.05)。结论缓脾1号方可改善STZ诱导糖尿病大鼠的糖脂代谢紊乱,其作用机制可能与抗氧化应激有关。
Objective To study the effects of Huanpifang Ⅰon glucose and lipid metabolism in STZ-induced diabetic rats and the study on the mechanism.Methods 60 male SD rats were randomly divided into 6 groups of 10 rats in each group,including blank group,model group,pioglitazone group,low dose group,middle dose group and high dose of Huanpifang Ⅰ group. Except the blank group,other groups were fed with high glucose and high fat diet combined with intraperitoneal injection of STC to establish diabetic rat model.Pioglitazone group was taken Pioglitazone 17.5 mg/(kg·d),the low-dose,middle-dose and high-dose groups were respectively given Huanpifang 10.3,0.7 and 1.4 g/(kg·d)respectively, while equal volume of normal saline was given once a day in blank group and model group. After 4 weeks,blood samples were taken to detect fasting plasma glucose(FPG),oral glucose tolerance test(OGTT),serum free fatty acid(FFA),total cholesterol(TC),triglycerides(TG),and low density lipoprotein cholesterol(LDL).ELISA was used to detect the activity of SOD and the content of MDA in liver. Liver histopathologic evaluation was performed by hermatoxylin-eosin staining.Results Compared with the model group,the level of OGTT was decreased significantly at fasting 30 minutes,60 minutes and 120 minutes in high dose group(P<0.01,P<0.05).The level of FFA,TC,TG and LDL was decreased significantly in high dose group and middle dose group(P<0.01,P<0.05). The activity of SOD was increased,and the content of MDA reduced in all dose groups(P<0.01,P<0.05). Conclusion Huanpifang I can improve the glyeolipid metabolism disorder in STZ-induced diabetic rats,which maybe due to anti-oxidative stress.
