摘要
目的建立巧克力豆中5种合成着色剂(柠檬黄、苋菜红、胭脂红、日落黄、亮蓝)及其铝色淀的高效液相色谱分析方法。方法样品用0.1%甲酸溶液洗涤,洗涤液经HLB固相柱萃取后采用高效液相色谱法进行测定。用XDB-C_(18)柱分离,以甲醇-乙酸铵(0.02 mol/L)为流动相,梯度洗脱,254 nm波长检测,外标法定量。结果柠檬黄、苋菜红、胭脂红、日落黄在0.5 mg/L~50 mg/L、亮蓝在2 mg/L~50 mg/L浓度内相关系数>0.999 5,柠檬黄检出限为0.2 mg/kg,苋菜红、胭脂红、日落黄检出限为0.5 mg/kg,亮蓝检出限为1.5 mg/kg。色淀样加标回收率良好,添加2 mg/kg、5 mg/kg、10 mg/kg 3个色素浓度水平的5种色淀标准,回收率在82.9%~112.0%,相对标准偏差在1.5%~6.2%。结论该方法简便、快速、灵敏、准确,适用于巧克力豆中5种合成着色剂及其铝色淀的测定。
Objective To develop a method for simultaneous detection of 5 synthetic colrants and their aluminum lakes in chocolate beans by high performance liquid chromatography(HPLC). Methods Chocolate beans samples were washed with 0.1% formic acid solution and purified by HLB solid-phase extraction column. The separation of 5 synthetic colorants and their aluminum lakes was performed on a XDB-C18 column using ammonium acetate solution(0.02 mol/L)-methanol as mobile phase. The samples were conducted for gradient elution, which were also detected by a UV detector and quantized by external standard with gradient elution method. Results Good linearity was observed when the concentrations of lemon yellow, amaranth, carmine, sunset yellow were within 0.5 mg/L-50 mg/L, and the concentration of bright blue was within 2 mg/L-50 mg/L, with correlation coefficient more than 0.999 5. The detection limit was 0.2 mg/kg for lemon yellow, 0.5 mg/kg for amaranth, carmine, sunset yellow, 1.5 mg/kg for bright blue. The recoveries of lakes at three concentration levels ranged from 82.9% to 112.0% for the 5 synthetic colorants and their aluminum lakes, and the RSDs were within 1.5%-6.2%. Conclusion The method is simple, rapid, sensitive, accurate, and it is suitable for the analysis of 5 synthetic colorants and their aluminum lakes in chocolate beans.
