摘要
随着生物治疗领域的发展,疗效确切、副作用小的生物大分子逐渐成为新药研发的热点,但是生物大分子作为候选药物在成药性方面存在一些技术难点,主要集中在药物稳定性和体内转运递送过程中,表现为生物大分子药物结构稳定性较差容易降解转化,生物大分子结构较大无法有效通过体内各种屏障系统进行递送等。为此,运用多样化的纳米载药系统对生物大分子药物进行递送,能够维持药物稳定性,显著增加生物利用度和提高分布靶向性,从而提高疗效并降低毒性。本文对目前一些生物大分子的纳米药物递送系统进行了介绍,并对其在相关疾病的研究进行了总结,比较全面地综述了包载生物大分子的纳米载药系统的研究现状和发展前景。
With the development of biotherapy, biomacromolecules with definite curative effects and fewer side effects have gradually become hot spots in the development of new drugs. However, as candidate drugs, there are some technical difficulties in the druggability of biomacromolecules, especially the drug delivery process in vivo. The structure of biomacromolecular drugs is unstable, easily decomposed and transformed in vivo. The biomacromolecules with large molecular mass can not effectively pass various barrier systems in vivo. The use of nano-drug delivery systems to deliver biomacromolecular drugs can prolong the half-life, increase the bioavailability and improve the distribution targeting, which greatly improves the efficacy of biomacromolecular drugs while reducing their toxicity. This paper summarized the biomacromolecules and nano-drug delivery systems as well as the researches of related diseases. The researches and development prospects of nano-drug delivery systems for biomacromolecules are reviewed.
