摘要
为探讨苏丹红Ⅰ(Sudan Ⅰ)对大鼠肝脏细胞色素P450酶系CYP 1A1、CYP 2E1和CYP 3A1基因表达的影响及ALT和AST活性的变化情况,将30只清洁级SD大鼠随机分为5组:对照组和Sudan Ⅰ处理组(包含四个剂量处理组),连续饲喂10 d,采集肝脏和血液作为检测样品。结果显示,Sudan Ⅰ处理组在一定剂量范围内,CYP 1A1、CYP 2E1和CYP 3A1基因的表达量呈上升的趋势,显著高于C组(P<0.01,P<0.05);ALT和AST的活性随着Sudan Ⅰ剂量的增加,呈现上升的趋势,且显著高于C组(P<0.05,P<0.01)。该结果提示,Sudan Ⅰ影响了CYP 1A1、CYP 2E1和CYP 3A1基因的表达以及ALT和AST的变化,从而发挥毒性,最终导致机体中毒,肝脏损伤。
In order to explore the effect of Sudan Ⅰ on expressions of CYP1A1, CYP2E1, and CYP3A1 genes and the concertration of ALT, AST in rats, experimental animals were randomly divided into five groups: control group and 4 Sudan Ⅰ treatment groups(There were four doses of the treatment group). All rats were killed and their liver and blood were collected for test at 10 days after treatment.The enzyme activity and blood components were measured to analyze the change. The experiment results indicated that CYP1A1, CYP2E1 and CYP3A1 genes were increased in a certain dose range(P<0.05, P<0.01), compared with control group. The concertrations of ALT and AST were increased in SudanⅠgroup(P<0.05, P<0.01). Concertrations of ALT, AST were significantly increased(P<0.05, P<0.01). These results indicated that Sudan Ⅰ affected the expressions of CYP1A1, CYP2E1, CYP3A1 and the activity of ALT, AST,implying that Sudan Ⅰ various kinds of poisoning effects on the organisms and injury of rat livers.
