摘要
目的研究羟喜树碱对人前列腺癌PC-3细胞增殖抑制作用及能量代谢和关键酶的影响。方法选取PC-3细胞,采用MTT法测定1.0、2.0及5.0μmol/L的羟喜树碱处理该细胞24 h后细胞的增殖抑制能力。采用分光光度法测定葡萄糖摄取量及乳酸生成量及ATP生成相关能量代谢参数。采用Western blot法测定乳酸脱氢酶A(LDHA)和AMP活化蛋白激酶(AMPK)表达。结果 1.0、2.0及5.0μmol/L的羟喜树碱可抑制PC-3细胞的增殖并呈剂量依赖性(P<0.05,P<0.01),IC_(50)值为3.24μmol/L。与对照组相比,不同浓度的羟喜树碱可降低葡萄糖摄取,抑制乳酸生成及ATP生成,且均呈剂量依赖性(P<0.05,P<0.01);Western blot结果显示,不同浓度的羟喜树碱可使PC-3细胞中LDHA及AMPK表达降低(P<0.05,P<0.01)。结论羟喜树碱通过可降低PC-3细胞中LDHA及AMPK表达,抑制肿瘤的有氧酵解并抑制细胞增殖。
Objective To investigate the effect of hydroxycamptothecin on proliferation and energy metabolism of human prostate cancer PC-3 cells and on the critical enzymes in these cells.Methods Using MTT method,the proliferation inhibitory ability was determined after the PC-3 cells were treated with hydroxycamptothecin at 1.0,2.0 and 5.0 μmol/L for 24 h.The energy metabolism parameters related to glucose uptake,lactic acid production and ATP production were determined with spectrophotometry.The expression of lactate dehydrogenase A(LDHA)and AMP-activated protein kinase(AMPK)was detected by Western blotting.Results Hydroxycamptothecin inhibited proliferation of the PC-3 cells at 1.0,2.0 and 5.0 μmol/L in a dose-dependent manner(P<0.05,P<0.01),with an IC50 value of 3.24 μmol/L.Compared with the control group,hydroxycamptothecin at different concentrations could reduce glucose uptake,inhibit lactic acid production and ATP production in a dose-dependent manner(P<0.05,P<0.01).Western blotting results showed that hydroxycamptothecin at different concentrations could reduce the expression of LDHA and AMPK in PC-3 cells(P<0.05,P<0.01).Conclusion Hydroxycamptothecin inhibits aerobic glycolysis and cell proliferation in the tumor by reducing the expression of LDHA and AMPK in PC-3 cells.
