大株红景天注射液维持心肌缺氧/复氧损伤的线粒体形态和功能的相关研究 被引量:5

摘要 目的研究大株红景天注射液维持心肌细胞缺氧/复氧线粒体的形态和功能。方法心肌细胞用流式细胞仪检测线粒体膜电位、线粒体通透孔,激光共聚焦显微镜观察细胞线粒体形态,并采用蛋白免疫印迹法检测p-Drp1蛋白的表达。结果大株红景天注射液能使缺氧/复氧心肌细胞线粒体膜电位升高,减少线粒体通透孔开放及线粒体分裂,抑制p-Drp1表达,实现线粒体损伤的保护作用。结论大株红景天注射液可减轻心肌细胞线粒体由缺氧/复氧引起的损伤,保护其形态和功能。 Objective To study mitochondrial morphology and function maintained by Sofren Injection after cardiomyocyte hypoxia/reoxygenation injury.Methods The mitochondrial membrane potential and mitochondrial permeability pore of cardiomyocytes were detected by using flow cytometer.The mitochondrial morphology was observed by using laser scanning confocal microscope(LSCM),and the expression of p-Drp1 protein was detected by using Western blotting assay.Results The mitochondrial membrane potential was increased,mitochondrial permeability pore opening and mitochondrial fission were decreased,p-Drp1 expression was inhibited,and mitochondrial injury was relieved by Sofren Injection after cardiomyocyte hypoxia/reoxygenation injury.Conclusion Sofren Injection can relive cardiomyocyte hypoxia/reoxygenation injury in cardiomyocytes,and protect mitochondrial morphology and function.
