青蒿琥酯对糖尿病大鼠牙龈组织炎症及IL-1β、TNF-α表达的影响 被引量:6

摘要 目的研究青蒿琥酯(ART)对糖尿病大鼠牙龈炎症及牙龈组织中IL-1β、TNF-α表达的影响。方法选取SD大鼠32只,随机分成对照组、ART50组、ART100组和正常组,其中对照组、ART50组和ART100组建立糖尿病模型后给予50mg/kg、100mg/kg ART灌胃处理4周。处死后获取标本,行HE染色及SABC免疫组化染色法检测牙龈组织中IL-1β、TNF-α的表达。结果 HE染色可见对照组大鼠牙龈上皮增厚,上皮钉突伸长,结缔组织排列紊乱,周围见大量炎性细胞浸润。ART50组、ART100组和正常组大鼠牙龈上皮角化明显,结缔组织内网状纤维排列整齐,少见炎性细胞浸润。对照组大鼠牙龈组织中IL-1β、TNF-α的表达高于ART50组、ART100组和正常组,差异具有统计学意义(P<0.05),而ART50组、ART100组和正常组之间差异无统计学意义(P>0.05)。结论糖尿病大鼠牙龈组织处于慢性炎症状态,50mg/kg、100mg/kg ART能抑制糖尿病大鼠牙龈组织IL-1β、TNF-α表达水平,发挥抗炎作用。 Objevtive To investigate the effects of artesunate (ART) on gingival inflammation and expression of IL-1β and TNF-α in gingivalof diabetic rats.Methods 32 SD rats were randomly divided into control group, ART50 group, ART100 group and normal group. The control group, ART50 group and ART100 group were established with diabetes model and administered with 50mg/kg and 100mg/kg ART for 4 weeks. Specimens were obtained after sacrifice. The expressions of IL-1β and TNF-α in gingival were detected by HE staining and SABC immunohistochemical staining.Results HE staining showed that the gingival epithelium was thickened, the epithelial spikes were elongated, the connective tissue was disordered, and a large number of inflammatory cells infiltrated around the control group. In the ART50 group, ART100 group and normal group, the gingival epithelial keratinization was obvious,and the reticular fibers in the connective tissue were arranged neatly, and inflammatory cell infiltration was rare.The expression of IL-1β and TNF-α in the gingival of the control group was higher than that in the ART50 group,ART100 group and normal group (P<0.05), but there was no significant difference between ART50 group,ART100 group and normal group (P>0.05). Conclusions The gingival of diabetic rats was in a chronic inflammatory state. 50mg/kg and 100mg/kg ART could inhibit the expression of IL-1β and TNF-α in gingival of diabetic rats and exerted anti-inflammatory effects.
